The development and biological characteristic in tumor relates to gene variation, which puts in action in protein translation. Therefore, the relation between eukaryotic protein translation initiation factors and tumor is being received much recognition, in which eIF4E (eukaryotic translation initiation factor 4E) is the most important one , in particular. eIF4E is an oncogene, which is a crucial rate-limiting modulator of protein translation initiation. Rencently, eIF4E was found to contribute to malignant transformation in cells.China has the first morbidity and mortality of eophageal carcinoma in the world, so exploring the mechanism of the invasion and metastasis of esophageal carcinoma has been one of research hot spots. However the relationship between eIF4E with the invasion and metastasis of esophageal carcinoma has not been reported. In order to discuss the mechanisms of the infiltration and metastasis of esophageal carcinoma, and extend survival time of esophageal carcinoma patients, cut down the mortality rate, Our research adopts immunohistochemistry S-P method, observing the expressions of eIF4E protein in esophageal carcinoma tissues of 58 cases, adjacent atypical hyperplasia tissues of 38 cases and normal mucosa tissues of 58 cases, illuminating furtherly the mechanisms of infiltration and metastasis of esophageal carcinoma and provides theory foundation for blocking invasion and metastasis of esophageal carcinoma.Methods1. Using the technology of immunohistochemistry SP to detect the expression of eIF4E in the 58 cases of normal mucosa of esophageal carcinoma, 38 cases of atypical hyperplasia tissues of tumor-adjacent and 58 cases of esophageal squamous cell carcinoma respectively.2. Statistical analysis: All the dates were analyzed by SPSS 10.0 statistical package, the count information calculated the positive rate, The comparison of positive rates uses the Chi-square. The level of significant difference isα=0.05.Results1. In 58 cases, the positive expression ratio of eIF4E in normal esophageal tissues, the tissues of atypical hyperplasia of tumor-adjacent and esophageal carcinoma tissues was 0%, 44.74%, 75.86%, respectively. The difference between three groups is remarkable.(P<0.05)2. In the 31 cases of metastatic group, the positive ratio of expression of eIF4E protein in the tumor tissues was 96.77%; in the non-metastatic group, the positive ratio of expression of eIF4E protein in the tumor tissues was 51.85%. The difference between the two groups was remarkable (P<0.05).3. The positive expression ratio of eIF4E in esophageal carcinoma tissues invade shallow muscle layer, deep muscle layer and chorion was 55.56%, 56.25% and 90.91%, respectively. The expression of eIF4E in the carcinoma tissues with chorion invaded is higher than that in the carcinoma tissue with shallow-muscle invaded and deep-muscle invaded. There was a significant difference among the groups(P<0.05). Nevertheless, the expression of eIF4E in the carcinoma tissues with deep-muscle invaded is compared that in with the carcinoma tissues with low-muscle, there is no significance (P>0.05).Conclusions 1. The positive expression ratio of eIF4E is different in normal esophageal tissues, the tissues of atypical hyperplasia of tumor-adjacent and esophageal carcinoma tissues. It is concluded that eIF4E participated in occurrence and development of squamous carcinoma of esophagus, which could be a marker for esophageal epithelium malignant transformation.2. The expression ratio of eIF4E in esophageal squamous cell carcinoma tissues with lymphnode metastasis is higher than that in without lymphnode metastasis, it indicates that there is correlation between the expression of eIF4E protein and metastasis of esophageal carcinoma.3. The positive expression ratio of eIF4E protein is different in esophageal squamous tissues with different invasion depth. The positive ratio of carcinomas tissues is increased gradually with the invasion depth advancing, and the positive ratio which invaded chorion is higher than that invaded shallow-muscle layer and deep-muscle layer. It indicates that there is correlation between the expression of eIF4E protein and invasion of esophageal squamous.
