Pyritum is one of the most important drugs used in orthopaedics departments. It must be processed before used. Calcining and quenching is used mostly to process pyritum at present. However, it has many weaknesses such as inefficiency, high intensity of working, polluting the environment, etc. So, we need to do something to solve these problems.We try to process pyritum by microbial leaching. We ground pyritum into power and sieved it. The we put them into 9 K liquid medium without adding FeSO4·7H2O or 9 K liquid medium without adding FeSO4·7H2O but with Acidithiobacillus ferrooxidans. Then we measured the lixivium regularly. And we found that there are more and more metallic ions in lixivium as time goes on. Thus, we can draw the conclusion that Acidithiobacillus ferrooxidans can oxidize and resolve pyritum power, and release metallic ions into the solution.Then we studied the effect of the leachate of pyritum on proliferation and differentiation of MC3T3-E1. We detected the cytoactive of MC3T3-E1 incubated with the leachate of pyritum by MTT. Hower, we didn't get the results what we had respected. The proliferation of MC3T3-E1 incubated with the leachate of pyritum dosen't show the relativity with concebtration of the leachate. And the proliferation of MC3T3-E1 incubated with the leachate of pyritum dosen't show the regularity that the proliferation of MC3T3-E1 increased or decreased as time goes on.Effect of pyritum leachate on union of fracture of Wistar bandicoots was observed. We administer leachate of pyritum to Wistar bandicoots by intragastric administration. The results are that alkaline phosphatase activities of blood serum of Wistar bandicoots which were administered by leachate of pyritum are higher than those of Wistar bandicoots which were not administered by leachate of pyritum. And the ion concentration of Ca2+, P of blood serum of Wistar bandicoots which were administered by leachate of pyritum is higher than those of Wistar bandicoots which were not administered by leachate of pyritum. The callus at the parts of fracture of Wistar bandicoots which were administered by leachate of pyritum is more than those of Wistar bandicoots which were not administered by leachate of pyritum. Thus, we can make a conclusion that leachate of pyritum can promote union of fracture of Wistar bandicoots by promoting sediment of the ion concentration of Ca and P and the formation of callus.
