To study whether anti sera against FSH was existed in the serum of infertility patients, we established the ELISA method for the detection of anti sera against FSH. The serum anti sera against FSH in some infertile or fertile men was detected. The result indicated that the incidence of the anti sera against FSH in the patients with oligospermia and azoospermia was significantly higher than that in fertility men. We presumed that the anti sera against FSH might affect the spermatogenesis of the male testis.To further research the effect of anti sera against FSH on male spermatogenesis, we established the rat model of anti sera against FSH. We had found that in model rats, the number of spermatogenic cells in seminiferous tubule and the sperm counts in tubular lumina decreased, when compared with control group. It confirmed our previous presumption that the anti sera against FSH might affect the spermatogenesis of the male testis.Based on the above experimental results, the serum hormones and the apoptosis of testicular cells were detected to explore the action mechanism of anti sera against FSH. The experimental results showed that the serum FSH level in model rat immunized with polypeptide for 77 d and 91 d was significantly lower than that in controls, However, the serum FSH level increased to normal in the model rat immunized with polypeptide for 105 d. Compared with the controls, the serum inhibin B level was significantly declined. The results of Fluorescent Quantitative PCR and TUNEL determination showed that the ratio of intratesticular Bax/Bcl-2 mRNA level, the mRNA level of caspase 3 and the apoptotic index of spermatogenic cells in model rat were all significantly higher than that in controls.Above all, we concluded that anti sera against FSH might cause the apoptosis of the spermatogenic cells and affect the rat testicular spermatogenesis. At the same while, the existence of anti sera against FSH for a long term might induce the establishment of a compensatory mechanism that could revival the repaired function of testicular spermatogenesis.
