The Laboratory Study Of The Effect Of Shen Yuan Dan(SYD)on Wistar Rats' Pharmaco Ischemic Preconditioning (PIP)

Ischemic preconditioning(IPC) has great effect of protecting heart, but it can't be used to clinic. So the reseach of pharmaco ischemic preconditioning (PIP) comes into being because of demand. The research we did before proved that Shen Yuan Dan(SYD) has the effect of counteracting myocardial ischemia , protecting endothelium, and accommodating neuroendocrine. The effect has relation of endothelin(ET), nitric oxide synthetase (NOS), angiotoninⅡ. This paper does deeply investigation of SYD's theory approach and its effect of pharmaco myocardial preservation and its mechanism.Theory studyThrough referring generous literature about PIP of chinese medical science and modern medicine , and basing its research progression, This paper reviews full-scale and systematically the research progression about PIP , the related record of traditional Chinese medicine (especially about ) . This paper observes the status quo of PIP brief and concise, quibus to unearth the related theory of traditional Chinese medicine about PIP.Laboratory studyObjectiveTo investigate the myocardial infarction area of Wistar rats'ischemia cardiac muscle , the change of nitric oxide synthetase (NOS) in blood serum, the expression of protein kinase C (PKC) and heat shock protein 70(HSP 70) , to approach whether SYD has the effect of PIP and how its mechanism reacted. Materials and MethodsResect 100 healthy ripe sanitation Wistar rats , equi- paroecious , weight 274±42 g ,about 4 months old , divided into 10 groups by radom: they are control group , reperfusion group , simulation ischemic preconditioning group , ischemic preconditioning group , drug and ischemic preconditioning group , drug andreperfusion group , simulation ischemic preconditioning/SWOP group , ischemic preconditioning/SWOP group , drug and ischemic preconditioning/SWOP group and drug and reperfusion/SWOP group , each group has 10 Wistar rats.. each rat was modeling, hemospasia, take their cardiac muscle. dipping; the cardiac muscle into 10% Formaldehyde, and made it dehydration , scaling wax, made section , staining.(1) Deal the sections with HE staining, under 4 times microscopic lens, analyze their area of myocardial infarction by color patho-picture analysis system software.(2) Extract the blood serum from the rats after centrifugalization, Investigate the vigor of NOS by ultraviolet spectrophotometry.(3) Deal with the a sections by PKC and HSP70 immunohistochemistry kit , obserbe the state of the expression of PKC and HSP 70 under 20 times microscopic lens.Results(1) SYD could diminish the area of the rats'myocardial infarction. Though SYD's effect isn't as good as IPC , they are synergism . It could strengthen the effect of diminish the area of the rats'myocardial infarction when use them together .(2) There is no significance difference of the vigor of NOS between each group , P﹥0.05.(3) PKC express obviously in ischemic preconditioning group , drug and ischemic preconditioning group and drug and reperfusion group , there's no expression in other groups . HSP70 express in drug and ischemic precon- ditioning group , drug and reperfusion group, ischemic pre condition- ing/SWOP group , drug and ischemic preconditioning/SWOP group , drug and reperfusion/SWOP group , others have no expression .Conclusions(1) SYD could diminish the area of the rats'myocardial infarction., it has the effect of protecting cardiac muscle as pharmaco ischemic preconditioning.(2) There's no relationship between NOS and the mechanism how SYD protect heart from ischemic attacking .(3) There do have some relationship between PKC and HSP70's mediating and the mechanism of the effect of SYD's pharmaco ischemic preconditioning.
