Abstract Aim: The synergistic anticancer effects of static magnetic fields(SMF) with cis-platin (DDP) on human leukemia cells K562 were studied. K562 cells were exposed under static magnetic fields and variation of Cyclin B expressing level was detected by western blotting.Methods: 1. The influence of SMF or/and DDP on K562 cells (human leukemia cells) were detected by MTT test. 2. After exposed under SMF with DDP, the morphologic modification of K562 cells were observed by light microscopy and atomic force microscopy (AFM); The cell cycle distribution of K562 cells were measured by flow cytometer(FCM); DNA damage of K562 cells were examined by Single cell gel electrophoresis(SCGE). 3. Variation of Cyclin B expressing level of synchronized K562 was detected by western blotting. 4. K562 cells were exposed under SMF, variation of Cyclin B1 level was detected by western blotting.Results:1.K562 cells with initial density of 1x10~5 cells/mL were exposed under SMF and DDP (10μg/mL) for 12h and growth of cells were inhibited.2. When K562 cells were treated with SMF and DDP(10μg/mL) for 12h, there were some debris of K562 cells by microscopy, there were some protuberances and holes-like structures appeared on the cell surface of K562 cells by AFM; the cell cycle distribution of K562 cells were measured by flow cytometer(FCM); the results showed that K562 cells can be arrested at the G2/M phase under exposure of SMF and can be blocked in the S phase by DDP treatment, but were mostly blocked S under exposure of SMF and DDP. The DNA damage was enhanced under exposure of SMF with DDP by SCGE; Changes in the structure of DNAunder exposure of SMF and DDP by AFM, found that cross-linking and fraction of DNA were increased.3. Variation of Cyclin B level of different cell cycle phase in K562 was detected by western blotting. Cyclin B began to express in early S phase, gradually increased in S phase, reached its peak level in G2/M phase, quickly disappeared in M phase late. 4. After exposed by SMF for 12h, 24h, 36h respectively, variation of Cyclin B1 level were detected with western blotting. The results showed under exposure of SMF, the level of Cyclin B1 has no significant changes compared with control.Conclusion: 1) The synergistic anticancer effects of static magnetic fields (SMF) with DDP on K562 cells were investigated. The modifications of cell morphology, cell proliferation inhibiting, DNA damage and the cell cycle distribution alteration might be the reason of anticancer effects by SMF and DDP. 2) After exposed by 9mT SMF for 12h, 24h, 36h respectively, variation of Cyclin B1 level were detected with western blotting. The result showed the level of Cyclin B1 has no significant changes under exposure of SMF.
