Objective To explore effects of Bone Mesenchymal Stem Cells (BMSCs)on healing of bone-tendon junction and provide experimental evidences on treatinginjuries of bone-tendon junction via Bone Mesenchymal Stem Cells (BMSCs)accelerating cellular proliferation and healing.Methods The BMSCs of rabbits were harvested by bone marrow aspiration.Adherent cells were selected as BMSCs after the whole marrow was cultured.Standard Animal model of partial patellectomy in 36 mature female New Zealandwhite rabbits (18-weeks-old) were established and randomly classified into 2 groups.The experimental group was planted into by the composite of BMSCs and PluronicF-127 while the control group was only operated without BMSCs planting into. TheQuadriceps Muscle-Patella-Patellar Tendon (QMPPT) were harvested at weeks 6, 12,18 postoperatively for gross, radiographic, histological (standard H&E stain,matrixproteoglycan and matrix collagen) and biomechanical examinations.Results Gross observations showed that the tissue healing in theexperimental group was quicker than that of controls. Radiographic measurementsshowed significantly more newly formed bone at the patellar tendon-patellar healingjunction in the experimental group compared with controls at weeks 6,12,18(P<0.01). Histologically, the experimental group at weeks 6, 12, 18 showed more improvedtissue integration, characterized by trabecular bone expansion from the remainingpatellar and regeneration of fibrocartilage zone at the patellar tendon-patellar healingjunction. Mechanical testing results(failure load and ultimate strength) proved it too(P<0.05). The histological observations and mechanical testing result showed thatthe tissue healing of the experimental group was quicker than that of the controlgroup.Conclusion BMSCs could accelerate celluar proliferation, matrix synthesis,new bone and fibrocartilage transitional zone formation, enhance mechanicalproperties, and accelerate bone-tendon junction healing.
