OBJECTIVE: To investigate the effect of the c-kit inhibitor Glivec on contraction of guinea-pig detrusor and the role of ICCs in contraction of guinea-pig detrusor.METHODS: The strips from detrusor smooth muscle were immersed into Krebs solution and linked with tonotransducer to recode their contraction. Glivec was delivered to the organ bath in the perfusing solution.RESULTS: Glivec decreased the amplitude of spontaneous contraction as compared to controll group(100μmol/L:547.87±16.70mg,p<0.01,n=15; 200μmol/L:359.65±17.70mg,p<0.01,n=15; 300μmol/L:197.47±15.20mg,p<0.01,n=15 for separate), descending degree of amplitude of detrusor smooth muscle contraction had minus correlation with Glivec concentration. There were no difference between controll group and 100μmol/L Glivec in frequency of spontaneous contraction( 100μmol/L:2.44±0.16次/分,p>0.05,n=15). Frequency of spontaneous contraction(200μmol/L:1.71±0.31次/分,p<0.01,n=15;300μmol/L: 1.11±0.68次/分,p<0.01,n=15 for separater)was lower than controll group. descending degree of frequency of spontaneous contraction had minus correlation with Glivec concentration.CONCLUSION: ICC-like cells has intimate relation with excitability and contraction of detrusor muscle, These results suggest that ICCs may evoke detrusor smooth muscle contraction as pacemakers. |
