Objective: To analyse the post articles shown the sorts of QingRe Chinese medicine have the function of anti-atherosclerosis ,and diabetes is a independent dangerous factor of atherosclerosis ,it act as a important role in the happening and developing of atherosclerosis, there are many diabetes patients accompany with atherosclerosis to be seen in clinical. We selected the sorts of QingRe Chinese medicine-DaHuang to study. To explore its protective mechanism in the vascular disease of diabetes mellitus, and provided a therapy access for diabetic atherosclerosis in clinical.Methods: 1 the parts of animal experiment: We randomly divided 50 Sprague-Dawley rats into control group (12 rats)and making model group(38 rats).The making model group rats were fasted for 12 hours, and then were injected Streptozocin(STZ) 60 mg/kg in intraperitoneal ,72 hours later ,took the blood from tail vein to measure the glycemia. We difineed the glycemia≥16.7mmol/l,and urine glucose+++~++++ for diabetes rats. After the diabetes rats model were finished ,choose 24 rats randomly,and they were randomly divided into Diabetes contrast group(12 rats)and DaHuang group.DaHuang group rats were lavaged with DaHuang water decoction 2ml,and the other two groups were lavaged with equal dose of pure water. During experiment ,rats were allowed to eat and drink freely,after the Chinese medicine–DaHuang was given 8 weeks constinously, rats were fasted for 12 hours ,took the blood from tail vein to measure the glycemia,and then were anesthetized, the blood specimen were collected from abdominal artoe , serum were checked for nitric oxide(NO), plasma were checked for endothelin-1(ET-1) by radioimmunoassay. After took blood from rats ,open chest cavity rapidly and cutted down the thoracic aortic ,each of thoracic aortic rings was cutted into 3mm length ,and each ring was hung in the vessel solt ,use the 10-6mol/l acetylcholine(Ach) to check the exactly endothelium, selected the rings of which could be inhibited more than 30% of rings constraction caused by 0.62×10-6mol/l norepinephrine(NE) to be experiment. Obseved the ratio of different concentration of Ach inhibited constraction caused by NE. At the time of took down the thoracic aortic ,and took down another part to make pathology slice,method of SP immunohistochemistry staining, using colorful image pathology anlysis system to calculated the positive ratio of intercellular adhension molecular-1(ICAM-1) and vascular cell adhension molecule-1(VCAM-1) between stained positive area and visual angel area. 2 The parts of clinical experiment : We selected 60 diabetes paitinets with constipation in clinical ,and randomly divided into three groups: Diabetes contrast group(20 paitients),TongBianLing group(20 paitients)and DaHuang group (20 paitients).After 1 month treatment ,took the blood and plasma were checked for ET-1 with the method of radioimmunoassay, serum were checked for NO with the method of nitratase. 3 Handled with statistics: All the datd were expressed withχ±? ,the software of statistics system was used with Statistical Package for Social Sciences 11.0(SPSS 11.0).One-Way ANOVA were used in compared of between groups,and Paired-Samples T Test were used in compared of before and after treatment of the group.Result: 1 the parts of animal experiment:①The glycemia was no significiant difference between before treatment and after treatment of Diabetes contrast group and DaHuang group (P=0.95),and there was no significiant difference between DaHuang group and Diabetes contrast group(P>0.05) ,there was no significant difference of glycemia in Control group between before lavaged and afer lavaged (P>0.05).②After 8 weeks ,Diabetes contrast group and DaHuang group comparied with Control group the plasma ET-1 was obviously increased (P<0.05﹚,and DaHuang group comparied with Diabetes contrast group the plasma ET-1 was obviously decreased (P<0.05﹚;The level of NO was obviously decreased in Diabetes contrast group and DaHuang group comparied with Control group(P<0.05﹚,and Diabetes contrast group comparied with DaHuang group was obviously decreased ( P < 0.05﹚.③Diabetes contrast group was obviously stronger than Control group and DaHuang group in response of NE caused constraction(P<0.05﹚,and also DaHuang group stronger than Control group(P<0.05﹚.After each group constracted to level groud ,then obey the ascendent method to given Ach (10-8~10-5) .The result show ,each group aortic rings show dose-dependent inhibited the constraction casued by NE.Under the same concentration of Ach , Diabetes contrast group and DaHuang group thoracic aortic rings response for Ach release effect obviously weaker than Control group (P<0.05). Diabetes contrast group thoracic aortic rings also weaker than DaHuang group (P<0.05).④The expression of ICAM-1 and VCAM-1 in rats thoracic aortic .There were only few brown-yellow granular positive staining substance expression in Control group rats ;And in Diabetes contrast group rats thoracic aortic rings there were lots of brown-yellow granular positive staining substance expression, parts of these substance mix into fragment,the positive staining ratio obviously higher than Control group (P<0.05);The brown-yellow granular positive staining substance expression in DaHuang group was lower than Diabetes contrast group (P<0.05),but higher than Control group (P<0.05).2.the parts of clinical experiment: There were no obviously changing of serum NO and plasma ET-1 in Diabetes contrast group and Tongbianling group before treatment and after treatment (P>0.05),and after 1 month treatment ,there were no obviously changing of plasma ET-1 and serum NO between Tongbianling group and Diabetes contrast group(P>0.05);The plasma ET-1 obviously decreased after treatment compared with before treatment in DaHuang group (P<0.05),and serum NO obviously increased (P<0.05).The plasma ET-1 of DaHuang group obviously decreased after treatment compared with the plasma ET-1 of Diabetes contrast group and Tongbianling group(P<0.05),and serum NO obviously increased (P<0.05).Conclusion: 1 There are dysfunction of the endothelia in these diabetic-rat models of this study through three months and six months. 2 The constraction of diabetic-rat blood vessel rings were stronger in induced of NE. 3 DaHuang could decrease the level of ET-1 and inhance the level of NO in diabetic rats , could protect the endothelia-dependent blood vessel relaxing function, and could inhibit the expreesion of ICAM-1 and VCAM-1 in aortic rings .4 DaHuang also could decrease the level of ET-1 and inhance the level of NO in diabetes patients in clinical experiment.This study show DaHuang through above mechanism to protect the blood vessel endothelia function, can inhibite the development of atherosclerosis,have the function of anti-atherosclerosis. 5 The function of DaHuang anti-atherosclerosis is multi-link, so there are wide exploiting and useing prospect. As the one of presendent of QingRe Chinese medicine, the function of its anti-atherosclerosis should be studied from multi-side and multi-level to carry on ,thus provide bases for clinical to use.
