Objective: To investigate the experimental effect on rabbit Masugi glomerulonephritis(MGN) treated by Shen-Le tablets and to analyze the possible therapeutic mechanism. The model of MGN was made by injecting sheep anti-rabbit kidney cortex serum to rabbit.And some indexes such as 24h urinary protein quantitative,plasma protein, blood-fat, renal function, renal pathomorphology, blood viscosity were observed, which will provid experimental data and evidence for clinical application of Shen-Le tablets .Method:Kidney cortex homogenate of rabbit mixed with complete freund's adjuvant(CFA) to suspl. Subcutaneouly injected the suspl to sheep with once every two weeks,4 times altogether. 2 weeks after the last injecton, took sheep blood, drew off the blood serum, mixed it with the same amount of rabbit erythrocyte ,and put it into the 4℃refrig for one night to ensure being fully adsorbted,took supernatant after centrifugali- zation, 56℃bain-marie to inactivate for 0.5h, and then sheep anti-rabbit kidney cortex serum obtained. Ordinary Newzealand big ear albino rabbits including 30 female and 30 androus were selected to our experiment.After adaptive feeding for one week, All rabbits were put into metabolism cage to obtain urine, and urine protein and red blood cells were detected. The result was negative. Then they were randomly divided into six groups: Normal group,Model group,TW group and low-dosage shen-le tablets group,middle-dosage shen-le tablets group, high-dosage shen-le tablets group (n=10,in each group). Anti-rabbit Kidney cortex serum was injected into each animal except the Normal group,2ml each time,once every 30 minutes,all 6 times. Obtain- ning positive reaction showed MGN model was successfully achieved. Drugs were administered to the rabbits of every treat- ment group and control group from the day after duplicating model but the distilled water were used in Normal and Model groups routinely. After model was made and the 28th day collected the rabbits'24h urine for 24h urinary protein quantitative assay. All animals were killed at the 29th day, throuth carotid artery cannula to get blood, serum TP, Alb, TC, TG,BUN,Scr were assayed with the automaticbiochemistry.Wh- ole blood viscocity, hematoplasma viscosity, fibrinogen and volume packed red blood cells were detected by blood viscosity instrument. Each group of nephridial tissue pathomorphology changes were observed with light microscope, electron microscope and immunofluorescence.Result: (1) 24h urinary protein quantitative:After model was made rabbits of all group showed a great quantity albuminuria except for Normal group, there was a significant difference compared with Normal group (P<0.01); At the 29th day the Model group urine protein quantitative compared with four treatment groups had the unusual significance difference except for low-dosage shen-le tablets group (P<0.01); though the low-dosage shen-le group urine protein is lower than the Model group, but there was no significant difference between them (P>0.05). High-dosage shen-le tablets group urine protein compared with TW group had no statistics significance (P>0.05).(2) biochemistry analysis: BUN and Scr of the blood serum in Model group were higher than Normal group significantly (P<0.01); BUN and Scr of other treatment groups descended significantly compared with Model group except for low-dosage shen-le tablets group (P<0.01);though BUN and Scr of the low-dosage shen-le tablets group is lower than the Model group, but there was no significant deviation between them (P>0.05). high-dosage shen-le tablets group BUN and Scr compared with TW group had no statistics significance (P>0.05).TP and Alb of the blood serum of Model group lower significantly than Norm- al group (P<0.01); TP and Alb of other treatment groups increased significantly compared with Model group except for low-dosage shen-le tablets group (P<0.01);though TP and Alb of the low-dosage shen-le tablets group is higher than the model group, but there was no significant difference between them (P>0.05);There was no significant difference between high-dos- age shen-le tablets groups and TW group (P>0.05). TC and TG of the blood serum of Model group increased significantly (P<0.01); TC and TG of the other treatment group is lower than Model group except for low-dosage shen-le tablets group (P<0.01,P<0.05); though TC and TG of the low-dosage shen-le tablets group is lower than the model group, but there was no significant difference between them (P>0.05);There was no significant difference between high-dosage shen-le tablets grou- p and TW group (P>0.05).(3)blood rheology analysis:ηb200s-1,ηb5s-1,HCTof Model group were higher than Normal group significantly(P<0.01);ηb200s-1,ηb5s-1,HCTof other treatment groups descented significantly compared with Model group except for low-dosage shen-le tablets group(P<0.01);thoughηb200s-1,ηb5s-1,HCTof the low-d- osage shen-le tablets group is lower than the model group, but there was no significant difference between them (P>0.05). There was a significant difference between high-dosage shen-le tablets groups and TW group (P<0.01, P<0.05).ηp200s-1,Fig of Model group increased significantly (P<0.01);ηp200s-1,Fig of the other treatment group descended compared with Model group except for low-dosage shen-le tablets group (P<0.01), thoughηp200s-1,Fig of the low-dosage shen-le tablets group is lower than the model group, but there was no significant difference between them (P>0.05);There was a significant difference between high-dosage shen-le tablets groups and TW group (P<0.01, P<0.05).(4)Pathomorphology of Nephridial tissue showed that: In the model group, the light microscope observation result showed that:glomerular basement membrane(GBM) was thickening,qua- ntity of intercapillary cells and mesenterium base increased significantly,a few of glomerulus manifested segmental glomer- ulosclerosis,cellular proliferation of glomerulus capsule wall,th- ere were crescent formation, proximal epithelial cell degenerated and swelling, there were proteinic cast in part of renal tubule lumens, inflammatory corpuscle infiltrated and scattered fibroplasias in the interstitial substance. Under electronic microscope segmental basal lamina was irregularly thickened,footprocesses of epithelial cells extensively confluen- ced and glomerular endothelial cells swelled seriously,which had remarkable difference compared with the high-dosage, middle-dosage shen-le tablets group and TW group.The low-dosage shen-le tablets group is homoioplastic with the model group.Renal tissue pathomorphology of TW group was similar to the high-dosage shen-le group.Immuunofluorescent microscopic examination showed that the fluorescent intensity reinforced markedly in Modle group,about ++~+++,and IgG deposited widespreadly in a"spot and liner"pattern along with GBM. The high-dosage, middle-dosage shen-le tablets group and TW group of the level of rabbit IgG deposited along with GBM were all less than Modle group,about+~++.Conclusion: 1We have succeed in setting rabbit Masugi nephritis models.2 Shen-Le tablets can help the rabbits with Masugi nephritis to decline proteinuria, increase the serum Alb and TP level ,alleviate blood hypercoagulability,decrease the rabbit's BUN and Cr to protect renal function and inhibit renal failure.3The pathologic changes indicated that the Shen-Le tablets can delay the progressin of interstitial fibrosis and glomerular sclerosis.4 Shen-Le tablets can possibly educe therapeutical effect throuth accommodating organism immune function,adjusting fatty substance metabolism,alleviating blood hypercoagulability,inhibiting inflammatory factor extemalizati- on and decreasing intercapillary cells proliferation.5 Shen-Le tablets may be similar to TW in the effect of treating glomerular nephritis,but in certain aspect possess ascendancy.
