Objective:The hepatic fibrosis of the liver was high occurrence rate and poor prognosis. The Traditional Chinese medicine has accumulated abundant experience of the treatment of hepatic fibrosis. According to the Traditional Chinese medicine theories and practice of curing this kind of disease, we developed this compound prescription Jiayijian. It also reflects Wang Xugao's and Qin Bowei's experience of using Chinese herbal medicine. According to the clinical application, the curative effect was remarkable. The aim of our experiment was to To investigate the effects of Jiayijian on the expression of MMP-2 and TGF-β1 in the rats with experimental DMN-induced hepatic fibrosis ,and to explore the possible mechanism of Jiayijian 's anti-hepatic-fibrosis.Methods: 60 Wister rats were randomly divided into the normal control and the model groups. The hepatic fibrosis model was induced in the experimental rats by intraperitoneal injection of DMN(dimethy nitrosamine) 10mg·kg -1·d -1 body weight ,three times a week for four weeks, half dosage on the first time. After hepatic fibrosis model being built, the rats in the model group were divided into the hepatic fibrosis model control group, the high, the low dosage Jiayijian. Experiment disposal : The rats in the Jiayijian groups were respectively given Jiayijian at the dosage of 22g/kg/d and 5.5g/kg/d,the rats in the normal control and the hepatic fibrosis model group were given the same volume saline, once a day for four weeks. All the rats were killed at the end of the ninth week. The severity of inflammation and fibrosis in all liver tissues were measured by semiquantitative scoring. At last, The expression of MMP-2 and TGF-β1 in liver tissues were detected by immunohistochemistryResults:1 compare the general state of rat health the normal control group growth well, weight gain, act agility the fur is very smooth, the normal urine and stool, the model group depressed and drowsiness, the fur is tangle and bloom, the yellow urine, and the unusual sedes, ,the treatment group weight is not decrease and act agility, the fur is no sheen, the urine and stool is okay.2 compare the liver tissue of the rats the liver of the normal control group looks bloom smooth and soft; the liver of the hepatic fibrosis model group looks little rough and the texture is crisp, the part of the liver is hard and lesser tubercle.3 compare the liver tissue under the microscope through the HE dyeing, we see the hepatic lobules construction are well in normal control group, liver cell cord is arranged on radiation around the central veins, the liver cells is arranged on multilayer mononuclear; pseudo-lobule being built in the hepatic fibrosis model group, liver cell cord is disorder, the fibrosis septa is increased and widen, the central veins are deviated or disappeared, cellular swelling and necrosis, the part atypical hyperplasia, the hepatic lobules of the high dosage yiayijian group are division partly, liver cell cord is arranged regular yet, there are small fabric and cell degeneration, or inflammatory cell in converge vessel field, pseudo-lobule is not completed in the low dosage yiayijian group, liver cell cord is disorder, fibroplasias and small inflammatory cell in converge vessel field. through the Masson dyeing: there are no collagen fibrils in normal control group, bulk of the collagen and lattice fibers appeared in converge vessel filed in the hepatic fibrosis model group, fiber septa is around of the pseudo-lobule, there are small green collagen fibers appeared in sinus hepatics and converge vessel filed in the high dosage jiayijian group, however,the green collagen fibers is more in the low dosage jiayijian group, and little then hepatic fibrosis model group.4 The score of inflammation activity and hepatic fibrosis Compared with the normal control group ,the severity of inflammation scoring and hepatic fibrosis scoring in the liver tissues of hepatic fibrosis model group was showed significant difference(P﹤0.01);Compared with the hepatic fibrosis model group, the severity of inflammation scoring and the liver fibrosis scoring in the liver tissues of all the Jiayijian groups became obviously lower, and the severity of inflammation scoring in the liver tissues of the high dosage Jiayijian group was markedly lowered, which showed significant difference (P<0.05).5 The expression of MMP-2 and TGF-β1 in liver tissues were detected by using in immunity class brood method.(1) Compared with the normal control group, the expression of MMP-2 in liver tissues of the other groups were obviously higher, which showed significant difference(P<0.01);Compared with the hepatic fibrosis model group, the expression of MMP-2 in the liver tissues of Jiayijian high, low dosage groups show no significant influence(P>0.05).(2)Compared with the normal control group, the expression of TGF-β1 in liver tissues of the other groups were obviously higher, which showed significant difference(P<0.01);Compared with the hepatic fibrosis model group, the expression of TGF-β1 of the rats in Jiayijian high, low dosage groups were lower, which showed significant difference (P<0.05).Conclusion:(1)Jiayijian may reduce the severity of inflammation scoring and hepatic fibrosis scoring in the rats of experimental DMN-induced hepatic fibrosis. (2)The expression of MMP-2,TGF-β1 in the liver tissues of the experimental hepatic fibrosis was enhanced, so MMP–2,TGF-β1 may be important factors in effecting hepatic fibrosis. (3)Jiayijian may down regulate the expression of TGF-β1 in the liver tissues of the experimental hepatic fibrosis, which may be the possible mechanism of Jiayijian 's anti-hepatic-fibrosis.
