Objective:NF-κB P65 is one of significant cell nuclear factors. Previous studies indicate that NF-κB P65 is over-expressed in most of human malignant tumors,but it can not be detected or weak-expressed in terminal differentiation normal tissue . NF-κB P65 becomes a new hot spot in researchs of carcinoma diagnosis and treatment because of its characteristic of relativity to tumor. Lung cancer is one of malignant tumors that usually occur in China. In the present study, the positive expression rates of NF-κB P65 protein, uPA protein,MMP9 protein and VEGF protein in non-small cell lung cancer were detected by immunohistochemical staining method. Meanwhile, we have analyzed the relation of NF-κB P65 protein to uPA protein,MMP9 protein and VEGF protein to explore the role of NF-κB P65 protein in the occurrence and advancement of non-small cell lung cancer and possible significance in diagnosis and treatment of tumor to provide a new method of assessing prognosis and targeting therapy.Methods: The samples of lung cancer tissues from 45 patients with non-small cell lung cancer were stained with polyclonal antibodies and monoclonal antibodies using Two-Step method. All the samples were pathologically diagnosed and surgically resected. The specimens from the same patient were obtained in normal lung tissues and lung cancer tissues. So the study included two groups,which were normal tissues group and lung cancer tissue group. Normal tissues was cutting edge surgically rescted tissue which was over 5 cm in size of mass. The results of immunohistochemistry staining were calculated according to cell stained intensity and the percentage of positively stained cells. Statistical analysis was performed by chisquare test and Wilcoxon rank sum test. P value less than 0.05 was considered to significantly different.Results1 The expression of NF-κB P65 in non-small cell lung cancer tissues and its relation to clinicopathological featuresThe positive expression rates of NF-κB P65 in non-small cell lung cancer tissues and nomal tissues were respectively 66.67%(30/45), 13.33%(6/45).There was significant difference between the two groups of patients(Mann-Whitney U= 422.500,Wilcoxon W=1457.500, Z=–5.412, P=0.000). The expressions of NF-κB P65 was highly associated with that of tumor size, differentiation of the tumor,pathological stages and lymph node metastasis(P﹤0.05).The expression of NF-κB P65 was not significantly correlated with other clinicopathological features such as sex, age, tumor spot and histology types(P﹥0.05). 2 The expressions of uPA,MMP9 and VEGF in non-small cell lung cancer tissues and its relation to clinicopathological features2.1 The positive expression rates of uPA in non-small cell lung cancer tissues and nomal tissues were respectively 80.00%(36/45),8.89%(4/45).There was significant difference between the two groups of patients(Mann-Whitney U=263.000, Wilcoxon W= 1298.000, Z= ?6.689, P=0.000). The expression of uPA was highly associated with that of tumor size, differentiation of the tumor and lymph node metastasis(P﹤0.05).The expression of uPA was not significantly correlated with other clinicopathological features such as sex, age, tumor spot and histology types and pathological stages (P﹥0.05).2.2 The positive expression rates of MMP9 in non-small cell lung cancer tissues and nomal tissues were respectively 73.33%(33/45),22.22%(10/45).There was significant difference between the two groups of patients(Mann-Whitney U= 465.500, Wilcoxon W=1500.500, Z= ?4.802, P=0.000). The expression of MMP9 was highly associated with that of pathological stages, differentiation of the tumor and lymph node metastasis(P﹤0.05).The expression of MMP9 was not significantly correlated with other clinicopathological features such as sex, age,tumor size, tumor spot and histology types (P﹥0.05).2.3 The positive expression rates of VEGF in non-small cell lung cancer tissues and nomal tissues were respectively 57.78%(26/45),20.00%(9/45).There was significant difference between the two groups of patients(Mann-Whitney U= 590.500, Wilcoxon W=1625.500, Z= ?3.896, P=0.000). The expression of VEGF was highly associated with that of differentiation of the tumor and lymph node metastasis(P﹤0.05).The expression of VEGF was not significantly correlated with other clinicopathological features such as sex, age, tumor size, tumor spot and histology types and pathological stages (P﹥0.05).3 The relation of NF-κB P65 to uPA , MMP9 and VEGF in non-small cell lung cancer tissues3.1 Spearman correlation analysis indicated that the expressions of NF-κB P65 and uPA were positive correlated in non-small cell lung cancer tissues( rs= 0.951 ,P=0.049).3.2 Spearman correlation analysis indicated that the expressions of NF-κB P65 and MMP9 were positive correlated in non-small cell lung cancer tissues( rs= 0.956 ,P=0.044).3.3 Spearman correlation analysis indicated that the expressions of NF-κB P65 and VEGF were positive correlated in non-small cell lung cancer tissues( rs= 0.951 ,P=0.049).Conclusions1 The expression level of NF-κB P65 is much higher in non-small cell lung cancer tissues than in nomal lung tissues. There is no significant relation between the expression of NF-κB P65 and age, sex, tumor spot,histology types (P>0.05) .There is positive correlation between the expression of NF-κB P65 and that of tumor size, differentiation of the tumor,pathological stages and lymph node metastasis(P<0.05).These results suggest that the expression of NF-κB P65 might play an important role in the occurrence, development, proliferation and metastasis of lung cancer. Therefore,it can be used as a useful marker for prognosis of lung cancer.2 The expression level of uPA is much higher in non-small cell lung cancer tissues than in nomal lung tissues. There is no significant relation between the expression of VEGF and age, sex, tumor size, tumor spot , histology types and pathological stages.There is positive correlation between the expression of VEGF and that of differentiation of the tumor and lymph node metastasis(P﹤0.05).These results suggest that the expression of VEGF might play an important role in the occurrence, development and metastasis of lung cancer.5 The expression levels of NF-κB P65 and uPA are high in non-small cell lung cancer.Meanwhile, there is significantly positive correlation between them.These results indicated that the target gene uPA might be upregulated by NF-κB P65 in non-small cell lung cancer .6 The expression levels of NF-κB P65 and MMP9 are high in non-small cell lung cancer.Meanwhile, there is significantly positive correlation between them.These results indicated that the target gene MMP9 might be upregulated by NF-κB P65 in non-small cell lung cancer .7 The expression levels of NF-κB P65 and VEGF are high in non-small cell lung cancer.Meanwhile, there is significantly positive correlation between them.These results indicated that the target gene VEGF might be upregulated by NF-κB P65 in non-small cell lung cancer .These results suggest that NF-κB P65 might participate in the formation of tumor micro blood vessels,and the soakage and transfer of lung cancer cells.Thus it may be employed as a clinicopathological marker of prognosis.
