Objectives To observe effect of sulphur dioxide(SO2)inhalation on the change of eosinophils infiltration and SP-D expression of mice nasal mucosa and on the change of IL-5, IL-13 in the peripheral serum and cavitas nasi irrigating solution,and explore toxic effect of SO2 on respiratory tract mucosa.Methods1.The animal modelsFifty male Kunming strain mice were randomly allocated to five groups:control group, allergic rhinitis group(AR group),and complex model groups(allergic rhinitis with three kinds of concentrations of SO2 groups).①AR group:mice were intraperitoneally injected with a mixture of OVA once every other day for seven times,and topically challenged with 5%OVA intranasally once every day for seven times.②Control group:the OVA was replaced by physiological saline in the same way.③Complex model groups:models were established by two steps.Firstly AR model groups were made in the same way.Secondly AR models were exposed to 56 mg/m3 SO2,112 mg/m3 SO2 and 168 mg/m3 SO2 respectively after topical challenge by the technique of dynamic fumigation once every day,4 hours every time,for seven days.2.The collection and analysis of samplesAfter establishing animal models,Cavitas nasi irrigating solution was gathered from cavitas nasi,and blood from the orbital venous sinus after anesthesia.The concentrations of IL-5 and IL-13 in the peripheral serum and cavitas nasi irrigating solution were measured by ELISA.The heads were embedded with paraffin,and serial sections were made and stained with hematoxlin-eosin.The number of eosinophils per square millimeter in sinus mucosa(Eos/mm) was calculated by using a computer-aided special software under microscope.The expression of SP-D in nasal mucosa was analyzed by immunohistochemistry technique.Results The concentrations of IL-5 and IL-13 in the peripheral serum and cavitas nasi irrigating solution in AR group were significantly higher than those in the control group(P<0.01).The density of sinus mucosa Eos in AR group was also significantly higher than that in the control group(P<0.01).The concentrations of IL-5 and IL-13 in the serum and cavitas nasi irrigating solution were no significant difference between 56mg/m3 SO2 group and AR group(P>0.05).The concentrations of IL-5 and IL-13 in the peripheral serum and cavitas nasi irrigating solution of 112mg/m3 SO2 and 168mg/m3 SO2 groups were significantly higher than those of AR group(P<0.01 or P<0.05).The density of sinus mucosa Eos in 112 mg/m3 SO2 and 168mg/m3 SO2 groups was significantly higher than that in AR group(P<0.01).There was positive correlation between the concentration of sulphur dioxide inhalation and the density of sinus mucosa Eos(r=0.887,P<0.01),and the concentrations of IL-5 and IL-13 in the peripheral serum(r=0.894 and 0.874,P<0.01),and concentrations of IL-5 and IL-13 in cavitas nasi irrigating solution(r=0.894 and 0.891,P<0.01).The expression of SP-D in56 mg/m3 SO2 and 112mg/m3 SO2 groups was all significantly higher than that in AR group(P<0.05,P<0.01).The expression of SP-D was obviously lower in 168mg/m3 SO2 group,and had no significant difference compared with that in AR group(P>0.05).ConclusionsThe concentrations of IL-5 and IL-13 in the peripheral serum and cavitas nasi irrigating solution and the density of nasal mucosa Eos of AR mice were significantly higher as sulphur dioxide inhalation.SP-D expression increased in 56mg/m3 SO2 group and 112mg/m3 SO2 group,but it decreased in 168mg/m3 SO2 group.The study showed that sulphur dioxide should aggravate the symptom of allergic rhinitis.
