Objective:To explore the expression of CD133 in human glioma cell of the patients with neurogliocytoma and to see whether or not CD133's expression associate with the differentiation and pathologic grading.It was expected that this study could reveal some essential qualities of the glioma and provide potential application for judging the malignant grades and biological treatment.Methods:The expression of CD133 and CD133mRNA was detected in 43 cases of human glioma and 16 specimens of normal brain tissues by hemi-quantitative reverse transcription polymerase chain reaction(RT-PCR)and Western-blot analysis,respectively.Results:①In human glioma samples,the positive rate of the CD133 mRNA and CD133 expression was 100%.In the control group,the positive rate of the CD133 mRNA was 6.25%, the expression of CD133 was negative.There were significance between two groups (P<0.01).②The gray degree ratios of the CD133 mRNA belt to theβ-actin mRNA belt varied accordingly to different pathologic grades.There were significant difference between gradeⅠand gradeⅢor gradeⅣ(P<0.01).The same significance was also found between gradeⅡand gradeⅢor gradeⅣ(P<0.01).The expressions of CD133 mRNA in gradeⅢor gradeⅣwere significantly more than those of gradeⅠor gradeⅡ.There were significant difference in the Reinhoit Zahl of the CD133 belt to theβ-actin belt among groups(all P<0.01)③Analyses of correlation:The positive correlation existed between the expressions of CD1331 or CD133mRNA2 and the pathologic grading of the human glioma(r1=0.987,r2=0.953,P<0.001).Conclusion:①Detecting CD133 in the human glioma can be used in diagnosing,judging the malignancy degree and the prognosis.②Detecting CD 133mRNA as molecular pathology in the human glioma would retrieve the limitations of traditional tissue pathology.③It will be an important cell marker of brain tumor stem cell that CD133 would act as more substantial role in future study.
