Object: To obeserve the effect of Akt genes on rats' liver fibrosis induced by CC14, and to investigate the anti-fibrosis mechanism of Akt genes in liver by mean of expressions of Bad and Caspase-9.Methods: Seventy male SD rats were randomly divided into group contrast(group C n=10), group liver fibrosis model (group L, n=20), group Akt-treated (group A, n=20) and group blank plasmid-treated (group B, n=20). No intervention studys were given to rats from group C. The rats from other groups were subcutaneously injected with 40% CCL4 eleo-solution in an amount of 0.5ml/100g, two times per week. Then we got some rats' hepatic tissue from group L in 8 weeks. Liposome-Akt and blank plasmid were respectively given to rats from group A and group B by mean of introportal infusion at the end of the sixth week, then got some rats' hepatic tissue at the end of eighth week. We compared the degree of hepatic fibrosis beyond the three groups according patho-grade, and detected the expression of Bad and Caspase-9 with immunohistochemistry, and analyze the difference of protein expression by mean of integral optical density.Results: 1 To successfully build rat model: the formatting rate of group L was 90%; group A was 85%; group B was 80%.2 we found the degree of rats' hepatic fibrosis in group A is more serious than that from group C, but better than that from group B and group L. According patho-grade of hepatic fibrosis, the grade of hepatic fibrosis from group A is significantly lower than that from group L and group B (P<0.01), but there is no difference between group L and group B.3 The expression of Bad and Caspase of rats from group L is significantly more than those from group C (P<0.01). And the expression of Bad and Caspase of rats from group A is significantly less than those from group L and group B(P<0.01), but there is no difference between group L and group B. Conclusion:1. Akt gene can inhibit rat liver fibrosis.2. The expression of Bad and Caspase-9 in hepatic fibrosis tissue are more than that in normal rat liver.3 Akt gene can inhibit rat liver fibrosis by inhibiting the expression of Bad and Caspase-9.
