Objective: To explore the association between G4A polymorphism of the Scavenger Receptor Class B Type I (SR-BI) gene and the occurrence of stroke and study the effect of G4A polymorphism on serum lipid level of Han population in Hunan province .Methods: G4A Polymorphisms of the SR-BI gene were detected by polymerase chain reaction(PCR) and restriction fragment length polymorphism (RFLP) in 149 atherosclerotic cerebral infarction patients (ACI), 141 cerebral hemorrhage patients (CH) and 120 healthy controls. Serum total cholesterol (TG) and triglyceride concentrations (TC) were mensurated by an enzymatic method. High-density lipoprotein cholesterol (HDL-C) was measured by an enzymatic method after heparin and calcium precipitated. Low-density lipoprotein cholesterol (LDL-C) was computed by the equation: (LDL-C) = (TC) - (HDL-C) - (TG/5). Results:1. The GA+AA genotype frequency and A allele frequency of SR-BIG4A in the ACI group, CH group and control group were 0.148 /0.074, 0.177/0.092 and 0.242/0.129 respectively. The A allele frequency in ACI group were remarkably lower than those in control group (P<0.05). The GA+AA genotype frequency and A allele frequency in CH group had no significant difference with those in control group(P>0.05).2. The serum level of HDL-C in SR-BIG4A GA+AA genotype subgroup was markedly higher than that in GG genotype subgroup of the same group in ACI group(P<0.05), and it was the same with CH group. The serum level of LDL-C in SR-BIG4A GA+AA genotype subgroup was evidently lower than that in GG genotype subgroup of the same group in ACI group(P<0.05), and it was the same with CH group. The serum level of HDL-C in SR-BIG4A GA+AA genotype subgroup was obviously higher than that in GG genotype subgroup of the same group in control group(P<0.05). The serum levels of LDL-C and TC in SR-BIG4A GA+AA genotype subgroup was significantly higher than those in GG genotype subgroup of the same group in control group(P<0.05).3. For male, the serum level of HDL-C in SR-BIG4A GA+AA genotype subgroup was markedly higher than that in GG genotype subgroup of the same group in ACI group and CH group (P<0.05). The serum level of LDL-C in SR-BIG4A GA+AA genotype subgroup was evidently lower than that in GG genotype subgroup of the same group in ACI group and CH group(P<0.05). The serum level of HDL-C in SR-BIG4A GA+AA genotype subgroup was obviously higher than that in GG genotype subgroup of the same group in control group(P<0.05). The serum levels of LDL-C and TC in SR-BIG4A GA+AA genotype subgroup was significantly higher than that in GG genotype subgroup in control group(P<0.05).Conclusions:1. The G4A polymorphism of SR-BI gene is a possible predisposing risk factor for atherosclerotic cerebral infarction. However, there is no association with cerebral hemorrhage of Han population in Hunan province. A allele is a possible protective factor for atherosclerotic cerebral infarction.2. A allele of SR-BIG4A is possibly associated with the metabolism of HDL and LDL of male.
