MTT assay was used to measure the level of the proliferation of Raji cells; the morphological changes of cells were observed by light microscope, fluorescence microscope and transmission electron microscope; the effect of matrine on cell apoptosis and cell cycle of Raji cells was studied by FCM. The expression of the survivin was measured with immunocytochemistry and image analysis. Results: in certain range of concentration, matrine could obviously refrain proliferation of Raji cells in a dose and time dependent manner; In result of fluorescence microscope and TEM, microvillus disappered, chromatin condensed to high electron density, nucleus broken to pieces and large deciduous apoptosis bodies appeared after being treated with matrine. The outcome of FCM showed that cell cycle was arrested at G0/G1 and G2/M, and matrine could introduce the apoptosis of Raji cells. The protein level of survivin was down regulated as the concentration of matrine was increased .Conclusion:1 Matrine could obviously inhibit the proliferation of human Burkitt's lymphoma Raji cells.2 Matrine could introduce apoptosis of Raji cells.3 Matrin could arrest the cell cycle at G0/G1 and G2/M phase.4 The expression of survivin was down regulated by matrine. | | Keywords/Search Tags: | matrine, lymphoma, Raji cell, apoptosis, cell cycle, survivin | PDF Full Text Request | Related items |
