Objective : To synthesize a new kind of chitosan-5-fluorouracil (5-Fu) nanoparticles and to study its characteristics of drug release and histocompatibility.Methods : The method of cross-liking ploymerid was used to synthesize CS-5-Fu NPs and detect their characterization by scanning electron microscope, AFM and PCS; the loaded capacity was nvestigated through ultraviolet spectrophotometry; high-performance liquid chromatography was used to detect the characteristics of intracorporal drug release about CS-5-Fu NPs. The CS-5-Fu NPs' toxicity was evaluated by MTT Method and in vivo histocompatibility experimentation.Results: The shape of CS-5-Fu-NPs was toroid and oblonga. Their dispersibility was fine. Their anteroposterior diameters were from 120~150 nm. The loaded capacity was (11.49±0.246) %. CS-5-Fu NPs as carrying agent had good histocompatibility and were atoxinogenic.Conclusions: CS-5-Fu NPs as carrying agent could change the behavior of intern pharmacokinetics and prolong the cycle time of 5-Fu in body. CS-5-Fu NPs could make initial burst reduced and the drug releasing rate was found decreased. CS-5-Fu NPs as carrying agent had good histocompatibility and were atoxinogenic. |
