The Effect Of The Different Form Of Jiawei Suanzaoren Tang On The Behavior Of The Rat And The Assay Of Ligustrazine,Sarsasapogenin In It

Objective: In order to probe the feasibility in application ofdispensing granule decoction, the study selected a famous Decoctionnamed Jiawei Suanzaoren Tang as experiment decoction, and comparedcomprehensively both the sleep and the autonomic activities of the ratand the contents of the different form of the decoction.Methods: 1. To compare the effect of the different form of JiaweiSuanzaoren Tang on the sleep of the rat, the study observed the effect ofthe threshold dose napental and the sub threshold dose napental on thesleep of the rat.2. To compare the effect of the different form of Jiawei SuanzaorenTang on the autonomic activities of the rat, the study observed the time ofaction and the frequency of the rat raising both feet.3. Using ultraviolet spectrophotometry to determine the content ofSarsasapogenin in the different form of preparation of Jiawei SuanzaorenTang.4. Using HPLC to determine the content of Ligustrazine in thedifferent form of preparation of Jiawei Suanzaoren Tang, C18 (4.6mm×250mm, 4μm) was used as chromatographic column; methanol- water asthe mobile phase=50:50(v: v), the column temperature 40℃, flow rate0.7ml/min, the detection wavelength 282nm; the sample size 40μl. Results: 1. In the test of the effect of the sub threshold dosesnapental on the sleep time of the rat, comparing the traditional slicedecoction and dispensing granule decoction of Jiawei Suanzaoren Tangwith the blank control group, the sleep time of the rat extended obviously;comparing respectively the traditional slice decoction and dispensinggranule decoction of Jiawei Suanzaoren Tang with the diazepam controlgroup, and comparing the traditional slice decoction with dispensinggranule decoction, there is no significant deviation. In the effect of thesubthreshold dose napental on the sleep number and the sleep time of therat, comparing the traditional slice decoction and dispensing granuledecoction of Jiawei Suanzaoren Tang with the blank control group, thenumber of the rat falling asleep increased and the sleep time of the ratextended obviously. But comparing the traditional slice decoction withdispensing granule decoction, there is no significant deviation. 2.Comparing the traditional slice decoction and dispensing granuledecoction of Jiawei Suanzaoren Tang with the blank control group, thetime of action and the frequency of raising both feet of the rat reducedobviously. But comparing each group, there is no significant deviation. 3.The linear relation of Sarsasapogenin was in the range of 8-65ug/ml, theregression equation was y=0.0122x+0.011,r=0.9992. 4. The linearrelation of Ligustrazine in the range of 2-45μg/ml, the regressionequation was y=37642x-14597, r=0.9991. Conclusion: Both the traditional slice decoction and dispensinggranule decoction of Jiawei Suanzaoren Tang have the significant effectof sedative and hypnosis(P＜0.05). It authenticate that the differentform of Jiawei Suanzaoren Tang both have the resemble effect of sedativeand hypnosis, which supply some experiment experience to the dosageform research of the traditional chinese medicine. The study determinethe content of Sarsasapogenin and Ligustrazine in the different form ofJiawei Suanzaoren Tang, which confirmed it can detect Sarsasapogeninand Ligustrazine in each form, and the content of Sarsasapogenin andLigustrazine in the dispensing granule decoction are both obviously morethan them in the tradition slice decoction. It confirmed that the extractionefficiency of dispensing granule decoction is complete, and affirmed thequality control of dispensing granule decoction.
