Objective: To explore the effects of bile acid on the cardiomyodial change of rat fetus.Method: Thirty clean SD female rats were randomly assigned into three groups after mating successfully. From day 13 to day 20 of gestation, Group A and B are injected intrapedtoneally with bile acid. The dose of group A is 5.5 mg.(kg.d)-1 and the dose of group B is 1.4 mg.(kg.d)-1. Group C was injected intrapedtoneally with Sodium Chlodde(NS). On day 21 of gestation, all rats were killed. TBA, DBIL, TBIL, ALT, AST and cTnI were measured in the maternal and fetal serum. Then the maternal and fetal hearts were collected for study. Pathohistological examinations of the hearts of the rat fetus were executed under light microscope and electronic microscope.Results: (1) Before intervening, the biochemical value in pregnant rat of every group shows no difference. (2) After intervened, the value of TBA in maternal and fetal serum of group A is (22.32±8.12)μmol/1, (28.84±8.06)μmol/1. Group B is (9.77±3.56)μmol/1, (9.34±3.54)μmol/1. Group C is (3.60±1.78)μmol/1, (3.95±1.19)μmol/1. The significant deviation exists between every two groups, P<0.01. (3)The fetal death rate in group A, B, C is 30.11%, 16.85%, 7.05%. The significant deviation exists between every two groups, P<0.05. (4)The value of cTnI in fetal serum of group A is (19.98±7.75) ng/ml. In group B it is (11.41±23.64) ng/ml. And in group C is (4.38±1.19) ng/ml. The significant deviation exists between every two groups, P<0.01. (5) Under light microscope, it showed that in the experiment groups abroad swelling, hemorrhage and necrosis occurred in myocardium tissue. Myocardocytes swelling seriously. The integrate of fetal necrosis area in group A is 1.92±0.43. In group B it is 1.36±0.37. In group C it is 0.44±0.12. The significant deviation exists between every two groups (P<0.05). (6) Under electronic microscope, it showed that in fetal rat cardiomyocytes of experiment groups, mitochonddon swelled, endocytoplasmic reticulum broke, myofibril ruptured, sarcomere blurred, cellular nucleus swelled and deformed. Under the electron microscope 20000 magnification, the number density and average velum of mitochondda in cardiamyocyte of group Ais (1753±206)μm-3 and (22.43±0.26)×10-3μm3. group B is (2672±182)μm-3 and (21.56±0.31)×10-3μm3. group C is (2428±269)μm-3 and (17.64±0.25)×10-3μm3. The number density and average volum of mitochondria in cardiamyocyte of group A are higher than those in group C (P<0.05) and the average volum in group B is higher than that in group C(P<0.05).The number density and average volum of myofibril in cardiamyocyte of group A is (1802±261)μm-3 and (7.51±0.26)×10-3μm3. group B is (2263±243)μm-3 and (14.59±0.34)×10-3μm3. group C is (2938±367)μm-3 and (12.64±0.25)×10-3μm3. The number density and average volum of mitochondria in cardiamyocyte of group A are lower than those in group C (P<0.05) and the number density in group B is lower than that in group C(P<0.05).(7) The positively correlation in maternal TBA, fetal TBA, fetal cTnI and the integrate of fetal necrosis area were found. The coefficient correlation between maternal and fetal TBA is r1=0.875(P<0.01). The coefficient correlation between maternal TBA ,fetal TBA and fetal cTnI is r2=0.540, r4=0.677(P<0.01); The coefficient correlation between maternal TBA ,fetal TBA and the integrate of fetal necrosis area is r3=0.675, r5=0.724(P<0.01); the coefficient correlation between fetal cTnI and the integrate of fetal necrosis area is r6=0.890(P<0.01). Conclusion: Fetal cardiomyocytes impared obviously in hypercholanemia. The serum level of TBA and cTnI in fetal rat were positively correlated with each other.
