IntroductionGastric stump cancer is a commom malignant tumour in the world. The incidence of gastric stump cancer shows a rising trendency. Accumulated evidence has established that it is associated with many gene.β-catenin and MMP-7 have been paid more attention to because of its key role in the initiation and development of carcinoma.β-catenin and MMP-7 were examined immunohistochemical S-P body staining method in 32 cases of human gastric stump cancer. The goal is to discuss the molecular mechanisms of gastric stump cancer and the possibility to discover in early stage, direct clinical and judge the prognosis.Materials and MethodsI .Material1. Samples: All sample used were obtained from paraffin -embedded tissues of the surgical speciments of patients admitted to the 1st Affiliated Hospital of China Medical University during 1998-2006.2. Reagents:β-catenin and MMP-7monoclonal antibody, S-P kit, DAB developer, 0.01mmolPBS, 3%H2O2, microtome, microscope.II. Methods1. staining:β-catenin and MMP-7 were examined immunohistochemical S-P body staining method in 32 cases of human gastric stump cancer. The dilute strength of MMP-7monoclonal antibody is 1:300.2. result assessment:β-catenin grade standard is on the basis of Jawhari and MMP-7 grade standard is on the basis of Yamamoto H.III. Statistical analysis Statistical analysis was performed using the t-test and Fisher correlation analysis, P<0.05 was considered statistically significant.ResultImmunohistoch- emical analysis demonstratedβ-catenin protein expression in 59.4% of the gastric stump cancer and in 12.5% of gastric stump mucosa (P <0.01) , whileβ-catenin expression was associated with macroscopic types, depth and size of tumors, invasive depth and growth pattern (P <0.05); MMP-7 protein expression in 56.3% of the gastric stump cancer and in 0% of gastric stump mucosa (P <0.01) ,while MMP-7 expression was associated with carcinoma invasive depth, growth pattern and lymph node metastasis (P <0.05). Theβ-catenin expression was in positive correlation with the expression of MMP-7 (P < 0.05).Conclusion1.β-catenin and MMP-7 are associated with developing, invasion and metastasis.2.β-catenin may enhance the degradative function of the target gene MMP-7 through nuclear translocation and may further facilitate local invasion and metastasis by the gastric stump cancer cells.
