objectiveIn this paper, to investigate the role of interleukin 6(IL-6) and matrixmetalloproteinases-9 (MMP-9) as well as the relationship of IL-6 and MMP-9 duringthe development of severe acute pancreatitis, we determined the expression of IL-6 andMMP-9 of pancreas tissue in a rat model of severe acute pancreatitis.MethodsSixty adult Wistar rats (170-190g) were randomly divided into six groups. Severeacute pancreatitis model on rats was established by retrograde injection of 4.5%sodiumtaurocholate (1mL/kg body) into biliopancreatic duct through duodenal wall. Rats weresacrificed at 1,2, 3, 4 and 5 days after induction of pancreatitis. The expression of IL-6and MMP-9 in pancreas tissue was determined by immunohistochemistry.Results(1)In control pancreas tissue, immunohistochemistry stain showed that IL-6 andMMP-9 expressions were absent and weak, respectively. The expressions of IL-6 andMMP-9 significantly increased in treatment groups, and were higher than those incontrol group (P<0.05). The expressions of IL-6 and MMP-9 reached their peaks at 3days and 4 days, respectively.(2)Correlation analysis revealed that IL-6 was positively correlated with MMP-9 (rs=0.317, P=0.025).Conclusion(1)The expressions of IL-6 and MMP-9 significantly increased in the SAP rats,which might play a key role in the development of SAP. The expression of IL-6 showedpositive correlation with that of MMP-9.(2)It revealed that the release of IL-6 during SAP might improve the release ofMMP-9 directly or indirectly.
