Object : To study the interaction of PPARγand EGFR signalling in Autosomal dominant polycystic kidney disease(ADPKD),for .Methods:1. To detect the changes of PPARγ, EGFR protein expression and EGFR mRNA in Han:SPRD rat renal tissues by immunohistochemistry,RT-PCR and Western Blot methods.2. To detect PPARγrelative activity through PPARγreporter plasmid FATP transient transfection to MDCK cell.Results:1. PPARγand EGFR overexpressed in ADPKD renal tissues. PPARγprotein was upregulated by EGFR tyrosine kinase inhibitor(TKI) . But EGFR protein and mRNA reduced when administered PPARγagonist.2. Protein expression and the activity of PPARγsignificantly reduced with EGF. But TKI can upregulate PPARγwith time prolonged and concentration stronger. Phosphorylation EGFR protein expression significantly reduced after given rosiglitazone with half an hour,though rosiglitazone had no significant effect on EGFR. Conclusions : PPARγand EGFR are co-overexpressive and inhibit each other in ADPKD. It was concluded that TKI may combinate with PPARγagonist to treat ADPKD.
