Objective To investigate the effect of bone marrow mesenchymal stem cell transplantation on vascular postangioplasty restenosis of rat. Methods 54 SD rats were randomized to be divided into control group, balloon injury group, balloon injury & MSCs transplantation group, MSCs were pre-labeled by DAPI, and then were infused into aorta through the balloon catheter (MSCs2×106animal). The piece of thoracic aorta were used for preparation of frozen section and paraffin section after 2,6 weeks respectively. MSCs labeled by DAPI were traced under immunofluorescence microscopy. The Histomorphology of aorta after HE staining was examined, and expression of smooth muscle alpha-actin(α-SM-actin) and proliferating cell nuclear antigen(PCNA) in aorta were analyzed with immunocytochemistry by using anti-αSMA antibody and anti-PCNA antibody. Results The DAPI labeled MSCs could be found on impaired intimae. Neointima formulation were found in injury group and transplantation group at 2 weeks. Expression ofα-SM-actin in the tunica media of aorta in transplantation group was significantly decreased and expression of PCNA in the neointima were obviously increased than injury group and control group at 2 weeks. Intima/tunica media area ratio and stenosis ratio of lumina in transplantation group were significantly increased than injury group at 6 weeks, p<0.05. Conclusion Bone marrow MSC transplantation could aggravate the vascular postangioplasty restenosis of rat, the mechanism maybe related with promoting the phenotype transition of SMCs in the tunica media and cellular proliferation in neointima after MSCs transplantation.
