Objective: To observe the effect of grape procyanidins (GPC) on the protein kinase C(PKC) expression in mice hepatic cells and to study the influence of PKC transductionpath for the proliferation of the hepatic cells.Methods: In vivo experiment: 40 BALB/C mice were divided randomly into control groupand 10,100 and 200 mg/Kg GPC group .The GPC solution was infused into the micestomach though mouth each day in GPC groups and distilled water was given in controlgroup for 30 days. The expression levels of PKC and PCNA protein in the hepatic cellwere measured by immunohistochemistry method. The proliferation of the hepatic cellswas determined by MTT assay, and the concentration of LDH in the hepatic cells weremeasured by chromatometry method. In vitro experiment: The hepatic cells werecultured with 10, 50, 100 mg/L GPC respectively for 48 hours. The expression levels ofPKC and PCNA protein in the hepatic cell were measured by immunocytochemistrymethod. The proliferation of the hepatic cells was determined by MTT assay, and theconcentration of LDH in the hepatic cells were measured by chromatometry method.Results1 The expression levels of PKC protein in the mice hepatic cellsIn vivo experiment: OD value of the high dose GPC group was higher than those ofthe low dose GPC group and control group(P<0.05). OD value of the middle dose GPCgroup were higher than that of control group(P<0.05). In vitro experiment: OD value of thehigh dose GPC group were higher than those of the low dose and the middle dose GPCgroup and control group(P<0.05).2 The expression levels of PCNA protein in the mice hepatic cellsIn vivo and vitro experiment: the OD value of PCNA protein expression in high doseGPC group was much higher than those of low dose GPC group and control group(P<0.05); the OD value of PCNA protein expression in the middle dose GPC group werehigher than that of control group (P<0.05)3 The proliferative activity of the mice hepatic cellsIn vivo experiment: The proliferation of mice hepatic cells in high dose GPC group was much higher than those of the middle dose GPC group ,low dose GPC group andcontrol group (P<0.05); the A value of the mid-dose GPC group were higher than that ofthe control group (P<0.05) .The rate of cell proliferation promotion in high dose GPCgroup was higher than that of low dose GPC group significantly(P<0.05). In vitroexperiment: The proliferation of mice hepatic cells from high to low: high dose GPCgroup ,middle dose GPC group ,low dose GPC group and control group (P<0.05). Thedifference of the cell proliferation promotion rate in high dose GPC group, low dose GPCgroup and the middle dose GPC group were significant in statistic (P<0.05).4 The concentration of LDH in the mice hepatic cellsIn vivo and vitro experiment: The concentration of LDH in high dose GPC group waslower than those of control group and low dose GPC group, and that of the middle doseGPC group was lower than control group(P<0.05).Conclusion:1 GPC can promote the proliferative activity of the mice hepatic cells and elevate theexpression level of PCNA protein.2 GPC can elevate the expression level of PKC protein and play important role forproliferation of the mice hepatic cells by PKC transduction.
