The Chronic Renal Failure(CRF) is a syndrome including a series symptom or metabolism disturbance that result in the renal function damage in progress cause by all idiopathy and secondary chronic renal disease. Now, the no-dialysis treatment on CRF including taking low- protein diet, controlling hypertension, et. And the treatment of TCM could put back the progress of Chronic Renal Failure, protect the renal function, improve clinical symptoms, raise the live standards, prolong lifetime.Shenweining Granule is a Chinese herb prescription on the treatment of chronic renal disease, it was elaborated by my supervisor professor Yu. It consists of Cordyceps Mycelium, Astragalus Membranaceus, Codonopsis Pilosula, Pericarpium Citri Reticulatae, Pinellia Ternate, Rhubarb, Leonurus Heterophyllu, Herba Ecliptae. It could invigorating Splleen and Kidney, clearing away Turbidness, removing blood stasis. Base on the nice effection of using Shenweining on clinincal practice, now we get going the animal experimental research. In the present study, we try to explore the possible effect mechanisms of Shenweining treating Chronic Renal Failure, give a estimation of availability and security of Shenweining treating CRF. And provide experimental evidence for the application of Shenweining further.Object:1. To explore the possible effect mechanisms of Shenweining granule.2. To give a estimation of availability about Shenweining granule treating Chronic Renal Failure.3. To evaluate the physic value of Shenweining and Coated AldehydeOxystarch by comparison. 4. To give a estimation of security about Shenweining granule treating Chronic Renal Failure.5. To evaluate the physic value of Shenweining using by different dose by comparison.Method:1. To choose the Wistar rats, to make animal models of Chronic Renal Failure by cutting 5/6 kidney according to the acknowledged criterion of internation.2. To disparted the Chronic Renal Failure rat models into 4 groups by randomized blocks: blankness group(Group A), lower dosage Shenweining group(Group B), higher dosage Shenweining group(Group C), Coated Aldehyde Oxystarch group(Group D).Every groups consists of 10 rats. Then give drugs by intragastric administration for eight weeks. To weigh them every week and observe their existent quality.3. To observe the existent state of all groups cases. To observe the physic value of all groups cases.4. To collect and record aria mount of 24h total urine of all groups cases whom have accepted therapy at the end of eighth week. To detect the levelof U-BUN, U-cr, Scr, BUN. To explore the possible effect mechanisms and availability of Shenweining.5. To take out the rest renal tissue specimen to make pathologic slice.Then observe their histological changes.Result:1. All rats of four groups were exist afer eight weeks for experiment. The weight of the lower dosage Shenweining group increased afer eight weeks experiment, (P<0.05), the weight of the rest groups were no significant difference. (P>0.05).2. All the Shenweining granule and Coated Aldehyde Oxystarch had decrease the level of Scr and BUN of rat models with 5/6 nephrectomy. The level of Scr in the higher dosage Shenweining group went down more than the lower dosage Shenweining group. The higher dosage Shenweining group showed the lowest level of Scr, the level of Scr were descend in the Coated Aldehyde Oxystarch group and the lower dosage Shenweining group. The level of Scr in the higher dosage Shenweining group and the Coated Aldehyde Oxystarch group and the lower dosage Shenweining group went down more significantly than the blankness group. (P<0.05, P<0.01). The level of Scr in the higher dosage Shenweining group went down more significantly than the lower dosage Shenweining group and the Coated Aldehyde Oxystarch group. (P<0.05). The level of Scr in the Coated Aldehyde Oxystarch group went down more significantly than the lower dosage Shenweining group. (P<0.05)。The level of BUN in the higher dosage Shenweining group and the Coated Aldehyde Oxystarch group went down more significantly than the blankness group. (P<0.01). The level of BUN in the Coated Aldehyde Oxystarch group went down more significantly than the lower dosage Shenweining group. (P<0.05).3. The amount of 24h total urine of the lower dosage Shenweining group was more than the higher dosage Shenweining group and the Coated Aldehyde Oxystarch group (P<0.05). The amount of 24h total urine of the rest groups were no significant difference. (P>0.05).4. The level of U-BUN in the higher dosage Shenweining group and the Coated Aldehyde Oxystarch group and the lower dosage Shenweining group increased more significantly than the blankness group.(P<0.01). The level of U-BUN in the lower dosage Shenweining group increased more significantly than the Coated Aldehyde Oxystarch group. (P<0.01). The level of U-BUN in the lower dosage Shenweining group increased more significantly than the higher dosage Shenweining group.5. The higher dosage Shenweining group showed the highest level of U-cr, the secondly high level of U-cr was showed in the Coated Aldehyde Oxystarch group. The level of U-cr in the higher dosage Shenweining group increased more significantly than the the blankness group and the lower dosage Shenweining group. (P<0.01). The level of U-BUN in the Coated Aldehyde Oxystarch group increased more significantly than the lower dosage Shenweining group. (P<0.05).6. Pathologic research showed:We had observed all pathologic slices of every groups. The pathologic slices of the higher dosage Shenweining group and the lower dosage Shenweining group showed more slight pathologic damage in renal tissue specimen. But the pathologic slices of the blankness group and the Coated Aldehyde Oxystarch group shoved more serious pathologic damage in renal tissue specimen.Conclusion:Shenweining granule and Coated Aldehyde Oxystarch could mitigate the damage in renal of rat models with 5/6 nephrectomy, and decreased the level of Scr and BUN, and improved the renal function, and protected the renal tissue, and could put back the progress of Chronic Renal Failure. The Shenweining granule is secure in treating the rat models with 5/6 nephrectomy. The curative effect of higher dosage Shenweining granule had better than the lower dosage Shenweining and Coated Aldehyde Oxystarch.
