Objective To make the reconstituted bone xenograft(RBX) whichcombine the mesenchymal stem cells(MSCs) and the os integumentale boneof BanNa inbred line small-ear pig. To observe the structuralcharacteristic of the composited bone. To explore the feasibility of thecomposited bone on repairing the macro-piece segmental radial bonedefects of the Japanese big-ear rabbits.Methods To make the 2.0cm macro-piece segmental radial bone defectsmodel of the Japanese big-ear rabbits. Transplant the the reconstitutedbone xenograft(RBX) which combine the mesenchymal stem cells(MSCs) andthe os integumentale bone of BanNa inbred line small-ear pig to themid-upper radial bone defects model of the Japanese big-ear rabbits. Theleft defects bones of the adult rabbits that as the experimental groupwere implanted into the composited bone which combined with themesenchymal stem cells (MSCs). The right defects bones as the control groupwere implanted into the uncombined xenogeneic bone. the defects were notfilled with anything in blank group. The repair capability of the defectswas assessed respectively after transplant operation by physical, X-ray,histology, bone mineral density examinations 4, 8, 12 and 16 weeks after operation. To compare the bone healing conditions of the three bonedefects groups.Results 16 weeks after operation, the bone defects were repairedcompletely and its bone mineral density is near to the normal bone levelin experimental group. The bone mineral density measurement after 4, 8, 12and 16 weeks postoperatively at all sites of bone defects revealed thatthe results of the experimental group were significantly higher than theircontrols. Comparing the experimental group, the defects were repairedslowly and the quantity of new bone formation is less in control group ,the defects of blank group were not repaired.Conclusion 1,Aftering drilling on the superficies externa wall ofthe os integumentale bone, the treatmented bone that origined the Inbredline small-ear pig have both the excellent structure and the goodbiomechanics intension. the treatmented bone which is the better choicein the cell bracket of the bone tissue project has the widely future inthe bone transplantation. 2,the MSCs which as the seed cells of the bonetissue project has the excellent superiority. 3,the MSCs compositedthoroughly with inbred line small-ear pig bone on repairing segmental bonedefects is obviously superior to the single xenogeneic bone in quantityand quality of new bone formation. the reconstituted bone xenograft hasthe good bone inducibility. 4,It is satisfaction result through the MSCscombined with inbred line small-ear pig os integumentale on repairingmacro-piece bone defects. 5,the reconstituted bone xenograft thatorigined the Inbred line small-ear pig is supplying a new and dependablemethod to treat the large bone defective.
