Background and ObjectiveAs one of the most common disease in the neurosurgery, brain injury has always been regarding as the emphasis in the neurosurgical field. At present, the incidence of brain injury is the second commonest in body injuries, but the incidence of mortality and impairment is the highest. With the development of transportation, reducing the incidence of brain injuries is the responsibility of the whole society and medicine. However, there is still large disparity between our country and the international in the treatment of brain injury. Such as inappropriate attention to the brain injury patients by the hospital staffs, lack of normalization and scientific principles in the treatment, poor ICU condition, and inadequate immediate and primary management of the disease. The inflammatory reaction of post-brain injury actually is organism's anti-damage reaction. How to take measures to decrease the damage degree of inflammatory reaction, control the intension, area, lasting period properly for excessive inflammation are the essential measures in the inflammatory reaction period of post-brain injury. This study tends to detect the changes of Interleukin-6 (IL-6) and C-reactive protein(CRP) in the serum of brain injury rat at the early-stage, to find the relationship between the changes and the degree of brain injury, and the prognosis as well.Materials and MethodsTotal of 30 adult Sprague-Dawley(S-D) rats weighing 250-300g (provided by the Animal Experiment Center of Zheng Zhou University) were divided into five groups randomly: the normal group, the sham-operated, the slight injury group, the injury group, and the severe injury group (six rats in each group) by using the method of Feeney's epidural strike to cause the injury. After the injury, blood was taken from rats' tails and serum IL-6 and CRP were detected by the method of enzyme-linked immunosorbent assay (ELISA), at 1h,12h,24h,36h,48h,72h respectively. All thedatas were recorded asx±s, and SPSS11.0 was used to analyze the data. Deviation and difference were compared using the ANOVA , t-test between two groups. The difference significanceα=0.05.ResultsAn acute elevation were observed in serum IL-6, and CRP levels in all the three groups divided by the degree of injury. The expression of IL-6 and CRP in severe injury group is higher than that in slight injury group and injury group(p<0.05) The expression was positively correlated with the degree of the injury(P<0.05).Conclusions(1) The level of IL-6 in serum of the post-head injury rises , especially in the severe injury group .And IL-6 takes an important part in inflammatory reaction of post-head injury.(2)The changes of CRP in serum of the post-head injury could reflect the degree of severity, especially in the severe injury group. CRP detection may become an independent factor to estimate the degree of severity in post-head injury.(3) The level of IL-6 and CRP in serum are important biochemical markers to estimate the degree of severity in brain injury. The expression was closely correlated with the severity of the brain injury, the severity and prognosis could be judged in time by observing the changes of IL-6 and CRP in serum.
