BackgroundThe brain stem reticular formation is a conformity center of the central nerve system. Nerve impulse is sent from this center to cerebral cortex, spinal cord, and other area to make a regulating function. The traditional reticular formation is looked as an old and dispersed area. There's just as no other nucleus except for a few clear nucleus. Except for splanchnic action (sutch as breath and circuitlation) regulation, it's just like no others in function. But with the much more research, there's other important and complicated functions of reticular formation. It's structure are more complicated than what we thought before. With the application of the immunofluorescence technique and immunohistochemical method, the new transmitter neurons were found. But there's still many questions for the brain stem reticular formation.The basic for the central nerve system is the structure. The come out nerve fibers from the reticular formation may cast to thalamencephalon, spinal cord, cerebella and other structures in brain stem. The fasciculus from the reticulat formation to spinal cord make a role in skeletal muscle regulation. At the same time, the neurons in the spinal cord can cast fibres to the reticular formation and regulate the accuracy of movement.To know more about the relationship between the reticular formation and the spinal cord neurons, we investigated the gigantocellular reticular nucleus and cervical spinal cord neurons.Methods1. The minim injection of WGA-HRP: the WGA-HRP was minim injected into the right gigantocellular reticular nucleus of grown rats. At the same time the negative comparison group was made.2. Detect WGA-HRP expression: 48h after the injection, the sections of the gigantocellular reticular nucleus and cervical spinal cord were subjected to TMB immunohistochemical reaction with ABC method. The result was detected and data was recorded and analysed.3. The minim injection of kinic acid: the kinic acid was minim injected into the right gigantocellular reticular nucleus of grown rats.This stimulation can bring c-fos expression in spinal cord neurons.4. Detect c-fos expression: 2h after the kinic acid injection, the c-fos expression was detected in cervical spinal cord neurons.Results1. The minim injection of WGA-HRP in the gigantocellular reticular nucleus can bring direct labeling in lamia I,II and recessive labeling in lamia VII,VIII of cervical spinal cord.2. The minim injection of kinic acid in the gigantocellular reticular nucleus can bring distinct expression of c-fos in lamia VII,VIII of cervical spinal cord. The expression was significant stronger than that in the negative control group (p<0.05).Conclusion1. The WGA-HRP and c-fos labeling indicate there's a direct ralationship between the gigantocellular reticular nucleus and cervical spinal cord neurons.2. The gigantocellular reticular nucleus cast fibres to the lamia I,II of cervical spinal cord and neurons in lamia VII,VIII of cervical spinal cord cast fibres to the gigantocellular reticular nucleus.
