Parkinson's diease named agitans paralysis , was a sort of nervous system degenerative disease, commonly onseted the middle-age and old people, which disease incidence reached 1% above 65 years crowd. Dyscinesia of the extracorticospinal tract was it's character,which was bradyinesia,statictremor,rigidity,autonomic neurofunctional disorder and aphrenia.Although previous cure method could relief Parkinsonian clinical symptom,which adverse reaction and complication were serious. Up to now,there was no radical cure and once onset,patients had to adopt cured all their time.For the reason, more researches about the reliable,effective, safe therapeutics were necessary in clinic and experiment, for solving patient's suffering .At present, various Parkinsonian animal models had established.The commonly used models were 6-OHDA rats PD model and MPTP Primate and mice PD model,which pathological change and ethology appearance was similar to mankind's PD character. They were reliable experimental models on studing Parkinsonian etiological factor, pathogenesy, drug treatment, neural graft and gene therapy .Acoording to large clinic experience and experiments,this task which was oriented on market and focused on patients, was selected Acanthopanacis senticosi as study object on the base of morning prescription study and gradually picked up the utility parts of Acanthopanacis senticosi roled on PD by pharmacodynamics experiments.There were the detailed experiment means as follow.1. To decide the optimum condition of PD mouse model induced by MPTP, by observing the effect on mouse ethology , substantia nigra of midbrain tyrosine hydroxylase(TH)and corpora striata dopamine (DA)content of different dosage MPTP. Consequence indicate all model groups could extend climb time, reduce TH~+ cell population and DA content on different degree.This result explained MPTP can induce mice PD symptom. According to the change and absence state of DA nerve cells and the decrease degree of DA content,the experiment choiced continuously injectted 5d MPTP , accumulative total count 150mg/kg,as the general standard of subac.model .2. To abserve mice ethology change , determine corpora striata MAO-B content, by the pharmacodynamics experiments on Acanthopanacis senticosi water decoct.and alcohol extracted liquor. Result indicated all the two groups liquor.could improve PD mice's motor coordination ability and MAO-B activity,which demonstrate the two groups liquor. have the therapeutic action. And the effect of water decoct. was notable.3. To use 70% ethanol precipitation on the water decoct. and get clear supernatant liquid and deposition ,then make the pharmacodynamics experiments on the two compositions separately.The result indicated, the clear supernatant liquid was active location.4. To make macroporous adsorptive resins column chromatogra-phy on the clear supernatant liquid,with water,30%,60%,90% alcohol eluting and get four elute groups,then make the pharmacodynamics experiments on the four compositions separately.The result indicated, 30% could improve PD mice's motor coordination ability an, degrade MAO-B activity and raise midbrain corpora striata DA content.There was no significant difference in the two groups ,on the climb experiment and MAO-B activity determination .So considered 30% and 60% alcohol eluting compositions as effective parts.
