Objective Attempt has been made to explore the role of celecoxib, a selective COX-2 inhibitor in the proliferation of human cervical cancer Hela cell line, and to investigate the relationship between celecoxib and the expression of cyclooxygenase-2,Ki-67 antigen.Methods The inhibitory effect of selective COX-2 inhibitor on the growth ofhuman cervical cancer Hela cell line was studied in vitro by MTT method. Flow cytometry(FCM) was used to analyze the effect of celecoxib on cell cycle parameters and apoptosis.The effects of celecoxib on the expression of cyclooxygenase-2,Ki-67 antigen were analyzedby immunohistochemistryResults MTT study showed that selective COX-2 inhibitor produced a dose-dependent and a time-dependent inhibition of Hela proliferation; FCM revealed that celecoxib could inhibit the growth of Hela cell line , influenced the cell cycle and induced the cell apoptosis. Immunohistochemistry revealed that celecoxib down-regulated the COX-2,Ki-67 expression of Hela cell.Conclusion Celecoxib could inhibit the growth of Hela cell line in vitro and the mechanism might be related to the influence of celecoxib in the cell cycle and the cell apoptosis induced by celecoxib...
