Dendritic cells are the most efficient antigen-presenting cells that powerfully activate T cells and initiate and sustain immune responses. Indeed, dendritic cells are able to efficiently capture antigens, express high levels of costimulatory molecules, and produce the combination of cytokines required to create a powerful immune response.Objective: To establish a stable culture system of dendrtic cell (DC) from peripheral blood monocyte cell in vitro, investigate the effect s of dendritic cell pulsed with tumor cell frozen-thawed antigen on inducing the cytotoxic T lymphocyte (CTL) to get specific anti-tumor activity in vitro.Methods: DC was generated f rom human Peripheral blood mononuclear cell (PBMC) in the presence of granuloceyte/ macrophage-colony stimulating factor ( GM-CSF) , interleukin-4 ( IL-4) in vitro. DCs were respectively pulsed with HCT-8, BGC-823 and SMMC-7721 frozen-thawed antigen ,and co-cultured to induce T cell into specific CTL. The effect s of specific tumor cell killing by CTL induced by DC pulsed with the tumor cell f rozen-thawed antigen and the effect of the changingof CD molecule expression on dendritric cells was observed.Results: The CD1a and CD83 expression on DC pulsed with frozen-thawed antigen increased more obviously than iDC( P < 0. 05), the extent of expression of CD1a increase more obviously than CD83( P < 0. 05). The CTL induced by DC pulsed with HCT-8 frozen-thawed antigen had more significant activity in HCT-8 cells killing than BGC-823 cell and SMMC-7721 cell in killing ( P < 0. 05). The CTL induced by DC pulsed with BGC-823frozen-thawed antigen had more significant activity in BGC-823cells killing than HCT-8 cell and SMMC-7721 cell in killing ( P < 0. 05). The CTL induced by DC pulsed with SMMC-7721 frozen-thawed antigen had more significant activity in SMMC-7721 cells killing than HCT-8 cell and BGC-823 cell in killing ( P < 0. 05).Conclusion: The CD1a and CD83 expression on DC pulsed with tumor frozen-thawed antigen increased. DC pulsed with tumor frozen-thawed antigen can induce the cytotoxic T lymphocyte (CTL) to get specific anti-tumor activity in intro but not unspecific anti-tumor activity.
