Objective: To discuss the effects of bone repairing between using Platelet-rich plasma and β -TCP recombination and using β-TCP only with the GBR in rabbits.Materials and methods: Twenty-seven New Zealand white rabbits, healthy, male, weight 2.5-3.0 kg. In each tibia, 10mm×5mm bone defect was created in the middle-upper part ( choose the joint between tibia and fibula as the middle marker ) One tibia was selected at randomly as experimental side, the other tibia was the control side. In the experimental side, the defects were filled with platelet-rich plasma and β -TCP, cover with collagen membrane then sew up each tissue layer. And in the control side, the defects were filled with β -TCP only, cover with collagen membrane then sew up each tissue layer. Each rabbit was injected with stretomycin in the following 3 days ( 250 thousand unite,muscle ) to prevent from imflamination. After 1 week, 3 weeks, 5 weeks, each rabbit was injected with fluorochrome under fur skin of its cervical part, (alizarin 30mg/kg, calcein10mg/kg, alizarin 30mg/kg ), After 4 weeks,8 weeks and 12 weeks, each time nine rabbits were killed, then all the tibias of the rabbits were taken out of and were divided into three different times experimental and control two groups. Then the tibia specimen were investigated by means of X-ray, histology of HE staining and microspecimen fluorescence microscope to compare the bone repairing.Results: Two rabbits died and other two were added. Bone grafts healed well without any infection in all cases. It was different between the two groups from histology of HE examination, after 4 weeks, the newly fiber tissues and cells in experimental group was more than in control group, and during the fiber tissue there are a few newly bone tissue; after 8 weeks, there are many bone tissue, little fiber tissue in the defect area in experimental group, but the defect area in control group has more fiber tissue than bone tissue, and the bone tissue is immature; after 12 weeks, the defect area was full of fibrous bone tissue in the experimental group, the defect area of the control group still has a few fiber tissue. The points of the newly bone density in X-ray examination: experimental group, four weeks 29. 35 ± 3.01 eight weeks 32. 35 ± 2. 53 twelve weeks 39. 79 ± 3. 36;control group, four weeks 13. 86 ± 2. 62 eight weeks 16. 68 ± 2. 61 twelve weeks 20.94 ± 3.29. The numerical value of area rate of newly bone tissue: experimental group, four weeks 42. 32 ± 1. 21 eight weeks 63. 91 ± 1. 31 twelve weeks 91.56±1.99; control group, four weeks 13.62 ± 2. 01 eight weeks 30. 96±1. 76 twelve weeks 60. 23 ± 1.43. And there was obviously statistical difference between the numerical value of two groups ( p < 0. 05 ) .Conclusion:1.PRP from the animal itself with bone substitution materials could improve the Osteo-Induction ability of the bone substitution materials, and it is synergic in conduction bone formation.2. Using of PRP with GBR could has better effect in the healing of bone defect.
