To reveal the effects of membrane surface charge on cell morphology, membrane elasticity and the structure and function of intracellular proteins of erythrocyte in normal physiological conditions, some advanced techniques like multi-dimensional micro-imaging technique, micro-imaging laser Raman spectral technique, dynamic micro-imaging analysis technique, and phase analysis electrophoretic laser-scattering technique were applied in this study. In addition, a micro-pipette aspiration technique was developed to measure the change of cell deformability analogue to the case of erythrocyte getting through capillary. The parameters of the structure and functions of single living erythrocyte, such as the morphology, membrane bending elastic modulus, Raman spectrum of intracellular Hb and membrane molecules, as well as the surface membrane Zeta potential, were determined and analyzed simultaneously. According to the experimental results, we have the following conclusion:1. Compared with young red cell, the old cell's membrane surface charge, membrane deformability, volume, ability of conveying oxygen decrease, while its concentration of Hemoglobin, the cell aggregation number increase.2. The membrane surface charge of old red cell can be recovered by adding Sialic acid(SA) ,Vitamin E(VE),N-acetylcystine (NAC). With membrane surface charge increased, the old cell's morphology, aggregation state, membrane deformability, and the hemoglobin's structure were comeback in different extent to the levels as the young one respectively. Among VE, NAC, SA, SA have the most powerful ability of modulate the concerned structural and functional parameters. In addition, SA improves the cell morphological and mechanical properties significantly.3. Membrane surface charge is not only the mark of whether the structural and functional parameters in normal physiological conditions, but may be also the key point to modulate the structural and functional parameters.This work not only reveals the correlation between red blood cell membrane surface charges and the cell morphology, membrane elasticity and intracellular protein,...
