Objective:The study is to investigate the bio-effects of VEC-304,SMMC-7721 cell line which were treated with low -frequency ultrasound combined with micro bubble agent .Methods:EVC-304 and SMMC-7721 cells are all divided in four groups : Group A: control ; Group B: pure micro bubble agent(made up of SB, Witamin C and HES);Group C: pure ultrasound; Group D: low-frequency ultrasound combined with micro bubble agent. The survival rates of VEC-304 and SMMC-7721 cell lines were observed by MTT; Apoptosis was analyzed by flow cytometry.Transmission electron microscope was used to detect the apoptosis and structure change of the cells. Visible spectrophotometric was applied to determine ROS and SOD . The expression of NF-κB was determined by Western blot.Results : The proliferation of Group D of VEC-304 or SMMC-7721 was obviously inhibited. The survival rate of VEC-304 and SMMC-7721 exposured 120swas 49±4.2%,51±3.5%, respectively, after treated 24h.There were significant differences between Group A and Group D. The survival rate of the cells, which were esposured 120s and treated 24h later, was the lowest; The apoptosis rate of group D was significant,which was reached (21.15±4.68)% in EVC-304 cell line and (27.31±4.14)%in SMMC-7721 cell line.There were significantly differences between Group A and Group D (P<0.01).The apoptosis rate of group C was lower than that of Group D, There were differences between GroupC and Group D(P <0.05).No differences of the between group A and B were found (P>0.05). Different degree morphological alteration had been found in the EVC-304 cell line of group D. It was presented karyopyknosis, chromatin gathering showed up clumping or crescent, apoptotic body, cytoplasm vacuolization and appear diffrent size cavity. cytoplasm disaggregation, chondriosome swelling,part of them had rupture of membrane, microvilli of the cell surface was reduced, even dissapear.There were similar changes in the group D of SMMC-7721 cells.
