ObjectiveBy applying polymerase chain reaction (PCR )to analyze the change of the four microsatellites sequences: D9S54, D9S162> D9S747,IFN—A1 in UCC urine and tissue, this research paper aims at exploring the relationship between MSI and biological behavior of bladder carcinoma, and in comparison with the current cytological examination seeking for the optimal simple but efficient approach to early diagnosis of bladder carcinoma and supervision of recurrence.MethodsThe samples of carcinoma tissue, peripheral blood and urine of 44 patients with the first attack, as well as 10 samples of these among healthy subjects free of carcinoma, were collected. Then these samples were stained with HE and diagnosed in the pathological department. In accordance with cystoscope coupled with CT, the carcinoma was graded with TNM; and it was pathologically graded according to WHO standards. 24 cases were found within the stage Ta-T1; 20 beyond the stage T2; 20 in the grade G1-G2; 24 in the grade G3. During the follow-up 3 to 16 months, the samples of urine and peripheral blood were extracted from 14 patients who were struck again.The samples above were tested with MSI. DNA samples were abstracted by the classical phenol~chloroform extraction method, and then these DNA samples were put into the 15μL, reaction system to be proliferated, and the proliferating products were electrophoresed with 1.8% gelose gelatin. Contrasted with the proliferating products from DNA of peripheral blood lymphocytes, the proliferating products from carcinoma tissue and urine samples were electrophoresed under the same condition.
