Objective:Stress is a state of the body response to any stimulus, and manifested by the activity of hypothalamo- pituitary- adrenocortical systems. Adrenocorticotropic hormone(ACTH) excreted by pituitary gland which is accommodated by corticotropin-releasing factor stimulates adrenal cortex to excrete glucocorticoids. Hematoplasma content of this hormone could result to metabolic changes of the body especially especial in brain tissues. So stress a state manifested by a series of neuroendocrine reaction and subsequent functional and metabolic changes of the body developed in response to any stimuli, especially enhanced activity of the sympathetic nervous system and hypothalamo- pituitary- adrenocortical systems.Glutamic Acid plays an important role in brain tissue injured by stress.In this study we give Propofol, Midazolam and ketamine to stress animal model to study expression of EAAT2,3, mGluR4,6and AKT,provide mechanism of Glutamic Acid and its recepter or transportor injure brain cells and Propofol protect brain tissue.Methods:1:The rat stress model was made by suspl crimp.The success of stress model was sure by observing the changes of the rat gastric mucosa.2: To observe the changes of rat brain tissue by HE dyeing.3: The expression of EAAT2,3,mGluR4,6 and AKTmRNA in rat brain was measured by RT-PCR. Results:The expression of EAAT-2,3 in stress rat group has significant increased in contrast with the control group. But decreased after giving Propofol, especial in 50mg/kg group,there are significant difference in contrast with stress group.In unstress group,the exoression of EAAT2,3 decreased after giving Propofol, it is provide that Propofol could decrease the expression of ETTA2,3. In Propofol group the exoression tendency of EAAT2,3 is difficient from Midazolam group,and idem with ketamine group. The expression tendency of mGluR4,6 in propofol group is idem with Midazolam group.The expression of AKT has no identical variation in each group,so we think that the variation of EAAT2,3 and mGlu-4,6 has no association with AKT.Conclusion :Propofol, Midazolam and ketamine could protect brain tissue ,and this effect has no association with AKT. The mechanism of Propofol is diffcient from Midazolam,but is resemble to ketamine.The tendency of mGluR in Propofol group is same with Midazolam group,so protection mechanism maybe has association with conscious-sedation effect of anesthetic .
