Objective : To study if Celecoxib have antitumor effect on glioma of rat in vitro, and investigate it's mechanism.Method: 9L glioma cell line and primary culture cell line of healthy rats were treated with Celecoxib at different concentration. effect of Celecoxib on cell growth was observed by methyl thiazolyl tetrazolium (MTT) method,Morphological Changes of 9L glioma cell apoptosis were observed by transmission electron microscope ,Hoechst 33258 staining and TUNEL;apoptosis rate was detected by Annexin - v-FITC/PI assay.Result :MTT method showed that Celecoxib at the concentration of 30~90μmol/L can inhabit the growth of 9L glioma cell line remarkably, while the effect on normal primary culture cell line is insignificant. After the treatment with Celecoxib,electron microscope, Hoechst 33258 staining and TUNEL were used ,both showed significant morphological changes in 9L glioma cells;apoptosis was observed by Annexin-v-FITC/PI assay, which showed that with increase of concentration of Celecoxib ,Apoptosis rate of 9Lglioma cell line was significantly elevated.Conclusion: Celecoxib can significantly inhibit the growth of 9Lglioma cell line in vitro, and it's main mechanism is induction of tumor cell apoptosis in dose-dependent manner. effect in Celecoxib -treated glioma cells is better than in Celecoxib -treated normal cells, which showed that Celecoxib have selective effect.
