Objective:To approach endothelialization and long-term patency rate of ePTFE (expanded polytera fluoro ethylene) and Dacron blood vessel prostheses with bone marrow CD34+cells.Methods:CD34+cells were isolated from bone marrow by canine and purified by immunomagnetic beads system, Seeding the cells to ePTFE and Dacron prostheses which implanted abdominal aorta artery(AAA) and inferior vena cava(IVC). Dogs were separated into ePTFE experimental group and ePTFE control group and the Dacron experimental group and Dacron control group, Obtaining the specimens of the experimental groups and the control groups to compare the patency rate in the 10,30,60,100 days after transplantation; biopsy specimens, and immunohistochemistry method identified endothelial cell; gely observed by light microscopy and scanning electron microscopy, and at different times of endometrial hyperplasia and endometrial endothelial, Observation of neointimal thickness; more arterial and venous system after bone marrow CD34+vascular endothelial cells seeded situation.Results:(1)The isolated cells from canine bone marrow were CD34+cells by flow cytometry. CD34+cells which isolated and purified by immunomagnetic beads system, trypan blue staining living cell to cell counting 2.6(±0.3)×10~7/ml. (2)Identification of CD34+cells by immunohistochemistry of the vascular wall cells growing endothelial cell layer.(3)Implanted arterial occlusive vascular endothelial artificial rate of 0, 25% stenosis group ePTFE Polyester Unit restenosis rate was 25%.
