Objective To investigate the expression of Protein Kiase B (P-AKT), Insulin-like growth factor-1(IGF-1), Vascular Endothelial Growth Factor(VEGF) and Microvascular Density(MVD) in non-small cell lung cancer and normal lung tissue and their correlations, to study the new diagnosed method of lung cancer through the relationship of P-AKT,IGF-1 and angiogenesis。Materials and Methods Expression of P-AKT,IGF-1,VEGF and MVD was detected by Streptavidin-Peroxidase (SP) method in 61 cases of NSCLC and 20 normal lung tissue respectively,the relationship between theirs expression and clinicopathological features of NSCLC was studied;at the same time,the relationship among them were also studied.Results (1) P-AKT protein was expressed in 27 of 61(44.3%) cases of lungcancer,significantly higher than that of normal tissue(0 of 20, 0%), (x~2= 13.28 P<0.05) . There was a significant correlation between P-AKT expression and differentiation,TNM stages and and lymph node metastases of NSCLC;(2)The MVD of P-AKT positive group was higher than that of negative group( 27.19±1.33 vs 23.83±3.06 t=6.61 P<0.05 ),and there was positive correlation between the expression of P-AKT and VEGF (r=0.38 P<0.05) .(3) IGF-1 was expressed in 42.6% (26/61) cases of lung cancer,higher than that of nomal tissues 10% (2/20) (x~2=7.09 P<0.05); the expression of IGF-1 was relative to lymph node metastasis ,TNM stages and differentiation in NSCLC;(4)The MVD of IGF-1 positive group was higher than that of negative group (27.04±1.46 vs 23.15±3.19 t=5.72 P<0.05) , and there was positive correlation between the expression of IGF-1 and VEGF ( r=0.287 P<0.05 ) . (5) The expression of P-AKT was relative to IGF-1 ( r=0.633 P<0.05) in NSCLC.(6)VEGF was expressed in 60.1 % (37/61 )cases of lung cancer ,higher than that of normal lesions 25% (5/20) (x~2=5.70 P<0.05) ,there was significant correlation between VEGF expression and lymph node metastasis ,TNM stages of NSCLC (P<0.05) ;(7)The expression of MVD in lung cancer and normal lung tissue was 25.04±3.11 and 19.37±5.20 respectively,obviously difference was found(t=5.913 P<0.05).Obvious correlation was found between lymph node metastasis , TNM stages and differentiation and MVD (P<0.05) .Conclusions (1)The expression of P-AKT and IGF-1 were correlation with lymph node metastasis , TNM stages and differentiation in non-small cell lung cancer,They participated in the happen and progress of non-small cell lung cancer and may also serve as definite diagnosis markers for non-small cell lung cancer.(2)There was positive correlation between the overexpression of P-AKT and IGF-1 and angiogenesis in NSCLC,they were both positive relation to the expression of VEGF.(3)There was positive correlation between the overexpression of P-AKT and IGF-1,IGF-1 could activate AKT.
