Objective To explored the correlation between the plamsa concentration of Bone Morphogenetic Protein-2(BMP-2) and the mumber oof Endothelial progenitor cells (EPC) in patients with idiopathic pulmonary arterial hypertension(IPAH).Materials and Methods We selected 28 patients with idiopathic pulmonary arterial hypertension (IPAH) and 20 matched control subjects, the plasma level of the bone morphogenetic protein-2 (BMP-2) was measured by ELISA.Total mononuclear cells (MNCs) were isolated from peripheral blood by Ficoll density gradient centrifugation, and then the cells were plated on fibronectin-coated culture dishes. After 7 days cultured, attached cells were cytochemical analysis. EPCs were characterized and counted as adherent cells double positive for DiLDL-uptake and lectin binding by direct fluorescent staining under a laser scanning confocal microscope.Results1. The mean levels of plasma BMP-2 were (0.0898 ±0.0295) ug/ml in normal controls and were (0.1294±0.0292) ug/ml in patients with IPAH( P<0.001) .2. The number of EPC was significantly reduced in subjects with IPAH compared with that of control subjects (31.8±7.7 vs 59.5±10.6 EPCs/x200 field; P<0.05) .3. The levels of plasma BMP-2 with IPAH was negatively correlated withthe number of EPC (r=-0. 736 P<0.001) . Conclusion1. The levels of plasma BMP-2 with IPAH were higher than that of control group , the number of EPC.was significantly reduced in subjects with IPAH compared with that of control subjects. These date indicated that he levels of plasma BMP-2 and the number of EPC were correlated with the degree severity of patient' s condition.BMP-2 may impair EPC-mediated reendothelialization after endothelial cell injury by inhibiting the number of EPC ,and BMP-2 and EPC were correlated with the degree of patient's condition and the clinical situation of IPAH disease2. The levels of plasma BMP-2 with IPAH was negatively correlated with the number of EPC, BMP-2 may impair EPC-mediated reendothelialization after endothelial cell injury by inhibiting the number of EPC ,and BMP-2 and EPC were correlated with the degree of patient' s condition and the clinical situation of IPAH disease...
