Reguralation Of Radix Astragali On Cell Signal Induced By HIF-1α In Cerebral Ischemia Damage Brains.

Objective To study the therapeutic effects of radix astragali injection(RA) on Insulin-like growth factor-1 (IGF-1 ),Hypoxia inducible factor-1α(HIF-1α) and heme-oxygenase-1 (HO-1) in rats brains following local cerebral ischemia/reperfusion. Methods One hundred eighty male adult SD rats were randomly divided into five groups: the sham-operated group, the model group,the low-dose RA group,the high -dose RA group and the Flos Carthami(FC) group. The model of middle cerebral artery occlusion(MCAO) was established by thread ligation method, and at 12h,30min before operation, rats were injected intraperitoneally corresponding medicine in different groups and the medicine was injected after operation every other 12h .In sham-operated group and the model group, equally dose saline was injected intraperitoneally. Neurological system symptoms were evaluated after 6h , 12h,24h,48h of ischemia/ reperfusion ,Then the rats were decapitated at 6h,12h,24h,and 48h after reperfusion,and rats brains were taken for immunohistochemistry and histopathological examinations, positive immune reacted cells of IGF-1, HIF-1α and HO-1 are counted under light microscope at different time points of ischemia/reperfusion.Results RA injection can significantly alleviate neurological system damage symptoms (p<0. 05 ) ;Histopathological examination showd that the cerebral ischemic damage in the RA group was significantly milder than that in the model group.and positive immune reacted cell amounts of IGF-1, HIF-1α and HO-1 were greatly enhanced in high-dose RA group as compared with the model group and the FC group (p<0. 05) .Conclusions The protective effects of the RA injection therapy against cerebral ischemia/reperfusion injury might be associated with improving the positive expressions of IGF-1, HIF-1α and HO-1 .Moreover,The therapeutic effects of high-dose RA is superior than the ones of FC and low-dose RA .
