Objective:To investigate Tetrandrine as a synergist to fluconazole in the yeast form and the hyphal form of Candida albicans in vitro and the mechanism.Materials and Methods:The minimum concentrations of Tetrandrine which are not fungistatic to the yeast form and the hyphal form of 16 C.albicans were confirmed according to microdilution test, and the minimum inhibitory concentration(MICs) of fluconazole alone and in combination with Tetrandrine were determined in 16 C. albicans strains by NCCLS M27-A or M38-P microdilution method. The percentage of the yeast form of 16 C. albicans containing Rhl23 were quantitatively assessed by flow cytometry assay(FCM). Fluorescence of Rhl23 in the cells were determined by fluorimeter. Results:When Tetrandrine ≤40 μg/mL, the survivability of the yeast form of 16 C. albicans was greater than 95%. Tetrandrine 30~40 μg/mL could restrain C.albicans from forming hyphae. When Tetrandrine ≤2 μg/mL, the survivability of the hyphal form of 16 C. albicans was greater than 95%.The MICs of fluconazole alone to the...
