OBJECTIVE:To observe the effect of ambroxol in chronic obstructive pulmonary disease (COPD) by establishing a model of COPD in rat by inhaling cigarrete.In order to provid the base of theory for the appliance of ambroxol ,the influence of it on animal model of COPD were studied by observing the change of ultra structures of bronchus, cell count and differentiation, and the level of interleukin-8 (IL-8 ) and tumor necrosis factor-α (TNF-α) in the bronchoalveolar lavage fluid(BALF).METHODS :Rats were randomly divided into three study groups: ① Normol control group (n=10); ② smoking Model group (n=10); ③ Ambroxol intervention group(n=10). The rat model of COPD has established by inhaling cigarette .For the ambroxol intervention group ,ambroxol was given 40 mg/kg daily for 60 days before the rats were put to death. The characteristics of ultrastructures of airway inflammation were analyzed by scanning electron microscope .The level of IL-8 and TNF-α' was detected by ELISA and the bronchoalveolar lavage fluid (BALF) cell count and differentiation were measured by Wright's staining.RESULTS: The model group manifested epithelial cell cilia of bronchial mucosa lossing, lodging and twisting, microvilli of chalice cell decreasing, inflammatory cell infiltrating, epithelial cell atrophing, chalice cell in region increasing , microvillus decresing. The surface of ciliated cell is thin and flat。 These feature correspond with the of human COPD.
