Low molecular weight heparin (LMWH) was obtained by chemical or enzymatic digestion of heparin. Its relative molecular weight was from 4000 Dalton to 6500 Dalton. LMWH was used to prevent deep phlebothrombosis after bone surgery, acute myocardial infarction, unstable angina pectoris, hematodialysis, extracorporeal circulation and so on. LMWH was administrated by injection although it was not convenient in the long-term injection because of its absorption difficulty through oral administration. Ethosomes is a new type drug carrier with high concentration of alcohol(20%~50%), which could form bilayer with better fluidity and deform easily. So, it could penetrate skin barrier to promote the drug absorption through skin. Here, we investigated the preparation and effects of the LMWH ethosomes, which was not studied before.The method to measure the envelopment rate of ethosomes was established. Then the components and preparation methods of the LMWH ethosomes were optimized through the experiment of four-factor three-level orthogonal design, with envelopment rate as measure standard. The essential components were LMWH 3%, phospholipid 10% and cholesterol 2. 5%, the envelopment rate was 39. 29%.
