Maidong is a Chinese traditional medicine widely used in China. It is the tubers of Ophiopogon japonicus (L.f.) Ker-Gawl. (Liliaceae). Chinese traditional medicine theory considers that the drug has the effects to nourish yin and promote the production of body fluid, to moisten the lung, and to ease the mind, being used to dry cough; phthisical cough; thirst due to impairment of body fluid; fidgetiness and insomnia; diabetes caused by internal heat; constipation.Column chromatography was carried out for the isolation and purification of the chemical constituents of tubers of O. japonicus. Two compounds were obtained, and their structures were elucidated by means of IR, Ms, 1H-NMR and 13C-NMR diffraction spectra date as ophiopogonin D (I) and ophiopogonin D' (II) . The purity of ophiopogonin D' was determined to be above 99%.An HPLC-ELSD method was established to determine ophiopogonin D' in tubers of O.japonicus. The contents varied from 0.0046% to 0.0107%. The method is simple, accurate, reliable and reproducible, and can be adopted for the quality control of tubers of O. japonicus.Fingerprint chromatography studies on methanol extracts of O. japonicus. were carried out by HPLC, and TCM fingerprint chromatography similarity evaluation system was used to evaluate the fingerprint chromatography of different market samples. The method established in this paper can be the reference to the quality study and evaluation of Tubers of O.japonicus.In order to study the distribution of heavy metals in O. japonicus. We detected the content of Pb, Cd, As, Cu and Hg in this plant by AA.
