| Objective: To investigate the effects of the combination of Progesterone and Mifepristone on proliferating inhibition and apoptosis in endometrial carcinoma cell lines HHUA.Methods:â‘ Endometrial carcinoma cell lines HHUA was cultured in vitro.â‘¡HHUA cells,treated by progesterone or mifepristone at different concentrations for 72 hours and the growth curve was drawn and then IC10 and IC50 respectively was calculated.â‘¢HHUA cells,treated by progesterone,mifepristone and combined two drugs at different concentrations(P IC10,RU486 IC10,P IC10+RU486 IC10,P IC50,RU486 IC50,P IC10+RU486 IC50,P IC50+RU486 IC10,P IC50+RU486 IC50)for 24h,36h and 48h were examined for their growth inhibition rates with MTT.â‘£HHUA cells,treated by progesterone and mifepristone in different groups(P IC10,RU486 IC10,P IC10+RU486 IC10,P IC50,RU486 IC50,P IC10+RU486IC10,PIC10+RU486IC10,PIC50+RU486IC50)for 48 hours the expression of Bcl-2 and Fas protein were determined by immunocytochemistry, Bcl-2 and Fas mRNA by RT-PCR.Results:â‘ The result of MTT showed that IC10 and IC50 of progesterone was 13.0547umol/l and 91.5584umol/l respectively,while IC10 and IC50 of mifepristone was 7.5604umol/l and 37.8352umol/l.â‘¡The result of MTT showed that progesterone combined mifepristone could effectively inhibit the growth of human endometrial carcinoma cell lines HHUA in vitro in time-dependent manner in 48 hours.(P<0.05)â‘¢Compared with that of control group,the expression of Bcl-2 and Bcl-2 mRNA of the combined group was significantly decreased and Fas and Fas mRNA was significantly increased(P<0.05).Conclusions: Progesterone and mifepristone can effectively inhibit the growth of human endometrial carcinoma cell lines HHUA in vitro in dose-dependent manner in 72 hours. Progesterone combined mifepristone can effectively inhibit the growth of human endometrial carcinoma cell lines HHUA in vitro in 48 hours.The combined group can reduce the expression of Bcl-2 and enhance the expression of Fas.These show that progesterone combined mifepristone can regulate the expression of Bcl-2 and Fas.The combined group may have some influence on growth inhibition and apoptosis of endometrial carcinoma,involving the levels of protein and gene. |
