Surface Plasmon Resonance (SPR) technology has been becoming a leading one for biomolecular interaction analysis because of its specific advantages. Biosensors based on SPR technology have been widely used in various fields such as biology, medicine, chemistry, and so on. SPR biosensor was a valuable tool for detecting kinetic biomolecule adsorption on material surface in real time and in situ. The amount of sample needed in analysis was small and without the necessity of radiolabelling technology. The concentration of biomolecule adsorbed on surface could also be determined according to the resonance angle shift.The aim of this thesis is to set up a SPR system which can be applied to evaluate the biocompatibility of biomaterials. This system which base on SPR technology can monitor the kinetic behavior of the protein adsorption onto the biomaterial surfaces in situ, in real-time and without labelling. The amount of proteins adsorbed on the biomaterial surfaces could be calculated according to the resonance angle shift and the blood compatibility could be evaluated preliminary.The main contents of this thesis are as follows:1. The research evolvement of biosensors based on SPR technology in the world was reviewed. The methods of biocompatibility and the protein adsorption were summarized briefly, and the development of the research on the biocompatibility by the SPR sensor was referred specially.2. The SPR principle was demonstrated theoretically, and the SPR-sensing configurations were discussed. Based on these, various factors to affect the SPR biosensor which based on the Kretschmann configuration were studied deeply, and the effecting results are simulated numerically by Matlab tool, which offers theoretical foundation for SPR biosensor's design and fabrication and debug.3. The hardware of a SPR biosensor which based on the Kretschmann configuration was rebuilt and optimized. The prism and the gold film were remade and data acquisition card based on the PCI bus was used for the Light Intensity Detection and Processing Unit.4. A set of software based on VC++ for the SPR system control and data acquisition, processing, analysis was designed with the hardware. Four functions and nine modules were designed in the software. And some debug and tests were done, and the SPR curves of Au in the atmosphere and the PBS were obtained. Compared the resonance angle of two SPR curves,the shift was obvious. According to the...
