Objective To research the feasibility of rapid Prenatal diagnosis of Down's syndrome by the Application of short tandem repeat PCR technique.Methods Based on the chromosome karyotype analysis as gold standard , amniotic fluid samplings from 51 Down's syndrome screening-positive mothers , peripheral blood DNA samples from 10 normal people and 10 Down's syndrome children .Using polymerase chain reaction to amplify four short tandem repeat sequence on chromosome 21:D21S1435, D21S11,D21S1270 and D21S1412 .The amplified products were analyzed using the PAGE electrophoresis and silver staining, the Down's syndrome was identified by the number and density of alleles.Results The technique of applying short tandem repeat-PCR on the prenatal diagnosis of Down's syndrome is established.
