Objective: To explore the infect of transferring and insulin on the proliferation of,human fibroblast in vitro and to establish the best cultivation method for harvesting considerable fibroblast quickly , and establish the experimental basis for clinical application.Methods: The fibroblasts from normal fresh skin were isolated and cultured and then subcultured to obtain sufficient experimental cells. Human skin fibroblasts were exposed to transferrin , insulin and both transferrin and insulin, MTT method was used for observing their proliferation in vitro.Result:1. A few funicular and bright fibroblast crawling out from the tissues were observed by inverted microscope in the 7th day of culture . Then the number of cell increased slowly, and adherent fibroblasts showed 2-5cell pseudopodia respectively .When the fibroblasts closed up on the bottom of culture bottle, they were distributed like stip and sheet , their nuclears were ellipse-like and small, and lied in the middle of cell. Many times after culture , the fibroblasts presented growth vigorous and HE stain showed from the 3rd generation most cells were fusiform-shaped .The 4th to 6th generation cells were used in our experiments.2.
