Background and Objective: The gastric hormone ghrelin has been reported to have a role in the regulation of bone metabolism on animals, and increase rat bone density, but whether it has the same function on human osteoblast remains unclear. In this study we investigated the plasma level of ghrelin in patients with osteoporosis(OP) and healthy adults, and evaluated the effect of ghrelin on the proliferation and on osteoblast activity in cultures of human periost osteoblasts.Methods: Serum ghrelin levels were measured in 17 patients with osteoporosis and 19 healthy adults. Human periost osteoblasts(POB) were cultured in vitro and ghrelin with different concentration was added to human osteoblast.The expression of alkaline phos,osteocalin,c and the promotion of calcium node were observed.Results: Serum ghrelin levels in patients with OP were lower than that in healthy controls. Ghrelin significantly stimulated osteoblast proliferation at the concentrations of 10-10mol to 10-9 mol, lower and higher concentrations had no effect on cell proliferation. 10-10mol ghrelin significantly increased the...
